摘要:
骨质疏松症是常见的全身性代谢性骨病,特征是骨量低、骨组织微结构破坏以及随之而来的骨脆性增加,易发生骨折。OP在我国发病率高居不下,严重影响患者健康生活,目前以药物防治为主。研究发现OP的发生发展与Wnt/β-连环蛋白(β-catenin)信号通路、Hedgehog信号通路、OPG/RANKL/RANK信号通路、磷脂酰肌醇3-激酶/蛋白激酶(PI3K/AKT)信号通路、核转录因子κB(NF-κB)信号通路等密切相关。中药单体、复方通过调控这些信号通路促进成骨细胞分化、抑制破骨细胞生成,改善骨代谢,增强骨密度,防治OP。本文旨在对OP与相关信号通路关系及中医药相关通路治疗机制进行探讨,为中医药防治的有效性、合理性提供科学依据,为临床防治提供新思路。
Abstract:
Osteoporosis was a common systemic metabolic bone disease characterized by destruction of bone mass,bone tissue microstructure,and subsequent increase in bone fragility and susceptibility to fracture.The incidence of OP remains high in China,which seriously affects health and life of patients.At present,treatment was mainly based on drug prevention and treatment.Studies have found that occurrence and development of OP were closely related to Wnt/β-catenin signaling pathway,Hedgehog signaling pathway,OPG/RANKL/RANK signaling pathway,phosphatidylinositol 3-kinase/protein kinase(PI3K/AKT)signaling pathway,nuclear transcription factor-κB(NF-κB)signaling pathway and other signaling pathways.Chinese medicine monomer and compound could promote osteoblast differentiation and inhibit osteoclast formation by regulating these signaling pathways,thereby improving bone metabolism and enhancing bone density to prevent OP.To review relationship between OP and related signaling pathways and mechanism of traditional Chinese medicine in the treatment of OP through related pathways,so as to provide scientific basis for effectiveness and rationality of traditional Chinese medicine in the prevention and treatment of OP.Provide new ideas for clinical prevention and treatment of OP.
