摘要:
目的:本研究以实验为基础,通过建立单侧输尿管梗阻(UUO)大鼠模型,对其形态学的影响及肾间质组织功能进行研究,并利用免疫组化实验方法,对其发生机制、抗肾间质纤维化的作用进行探讨。方法:本实验选取研究对象为60只SPF级健康雄性SD大鼠,随机分为手术组45只,假手术组15只,手术组大鼠在做完UUO模型后,再随机分成三组,模型组、苁蓉复肾方组、厄贝沙坦组,每组各15只。药液灌胃14 d后,各组随机选取10只大鼠,分别采集24 h尿液、血清和大鼠梗阻侧肾组织,用于测定血清肌酐、尿素氮、Ang-Ⅱ蛋白、MCP-1蛋白以及24 h尿蛋白等,其中梗阻侧肾组织一部分用于病理观察,另一部分用于免疫检测。结果:在苁蓉复肾方中的测定数值中,各剂量药组大鼠的血清肌酐、尿素氮比模型组大鼠明显降低。苁蓉复肾方组大鼠肾脏组织发生的病理变化,比模型组大鼠程度低。苁蓉复肾方组大鼠血清中Ang-Ⅱ蛋白的含量较低。大鼠血清中MCP-1蛋白在苁蓉复肾方组中含量较高。苁蓉复肾方组大鼠体内TNF-α蛋白与模型组相比表达量较低。苁蓉复肾方组大鼠中ColⅣ蛋白的表达低于模型组。结论:苁蓉复肾方可使血肌酐、尿素氮、24 h尿蛋白定量降低,减轻单侧输尿管梗阻引起的肾脏损伤。降低UUO模型大鼠的肾脏病理损害,促进其血清中MCP-1蛋白的表达,进而抑制血清中的Ang-Ⅱ蛋白、TNF-α蛋白,以达到降低ColⅣ的蛋白含量,减少ECM的合成和积累的目的,缓解肾间质纤维化进程。苁蓉复肾方在一定程度上保护肾功能,使肾小管损伤程度减轻,肾间质纤维化程度降低,为肾间质纤维化的临床治疗提供依据。
Abstract:
Objective:In this study,a rat model of unilateral ureteral obstruction(UUO)was established based on the experimental design to investigate its morphological effects and renal interstitial tissue function.Immunohistochemical methods were employed to explore the pathogenesis and mechanisms underlying anti-renal interstitial fibrosis.Methods:In this experiment,60 healthy male SD white rats of SPF grade were selected as research subjects,and they were randomly divided into surgery group(45),sham surgery group(15),and the surgery group rats were divided into three groups after the UUO model was made,namely,model group,Astragalus membranaceus Congrong Fushen prescription group,and Irbesartan group,with 15 rats in each group.After 14 days of oral administration of the solution,10 rats were randomly selected from each group to collect 24-hour urine,serum,and the obstructed kidney tissue of the rats for the determination of serum creatinine,urea nitrogen,Ang-Ⅱ protein,MCP-1 protein,and 24-hour urine protein,and part of the obstructed kidney tissue was used for pathological observation,and the other part was used for immune detection.Results:The Congrong Fushen prescription group exhibited significantly lower levels of serum creatinine and urea nitrogen compared to the model group.The pathological changes in the renal tissue of the Congrong Fushen prescription group were less severe than those in the model group.The Congrong Fushen prescription group also showed lower levels of serum Ang-Ⅱ protein and higher levels of serum MCP-1 protein.Additionally,the expression of TNF-α protein in the Congrong Fushen prescription group was lower compared to the model group.The expression of Col Ⅳ protein in the Congrong Fushen prescription group was also lower than that in the model group.Conclusion:Congrong Fushen prescription has a protective effect on renal injury induced by unilateral ureteral obstruction(UUO),reducing blood creatinine,urea nitrogen,and 24-hour urinary protein levels.It alleviates renal pathological damage in UUO model rats,promotes the expression of MCP-1 protein in the serum of these rats,and inhibits the expression of Ang-Ⅱ and TNF-α proteins in the serum.Consequently,it decreases the protein content of ColⅣ,reduces the synthesis and accumulation of extracellular matrix(ECM),and slows down the progression of renal interstitial fibrosis.To a certain extent,Congrong Fushen prescription protects renal function,mitigating the degree of tubular damage and reducing the extent of renal interstitial fibrosis,thereby providing a basis for the clinical treatment of renal interstitial fibrosis.
