摘要:
目的:探究针刺对失眠大鼠松果体凋亡相关蛋白的作用。方法:将48只SPF级雄性SD大鼠随机分为空白组、模型组、假针刺组、针刺组。除空白组外,其余各组通过腹腔注射对氯苯丙氨酸(PCPA)构建失眠模型大鼠。空白组不予治疗,模型组仅捆绑不治疗,针刺组予以针刺“百会”“神门”(双侧)、“三阴交”(双侧)干预,假针刺组在相同穴位予以假针刺治疗。借助戊巴比妥钠翻正实验测定造模后及干预后大鼠睡眠潜伏期及睡眠时间,运用RT-PCT检测松果体B细胞淋巴瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)、褪黑素(MT) mRNA表达,采用免疫组化检测大鼠松果体Bcl-2、Bax蛋白阳性表达。结果:造模后,失眠大鼠睡眠潜伏期延长,睡眠时间大幅缩短(均P<0.01)。治疗后,针刺组睡眠潜伏期缩短,睡眠时间延长(均P<0.01)。与空白组相比,模型组松果体MT mRNA表达降低,Bcl-2蛋白及mRNA表达下降,Bax蛋白及mRNA表达增加(均P<0.05)。与模型组相比,针刺组松果体MT mRNA表达显著升高,Bcl-2蛋白及mRNA表达升高,Bax蛋白及mRNA降低(均P<0.05),假针刺组无统计学差异(P>0.05)。结论:针刺通过调节失眠大鼠松果体凋亡相关蛋白表达,进而促进松果体MT分泌,缩短PCPA所致失眠大鼠的睡眠潜伏期,延长睡眠时间,改善睡眠状态。
Abstract:
Objective:To explore effect of acupuncture on apoptosis-related proteins in the pineal gland of insomnia rats.Methods:48 SPF male SD rats were randomly divided into blank group,model group,sham acupuncture group and acupuncture group.Except for the blank group,the other groups were given insomnia model rats by intraperitoneal injection of p-chlorophenylalanine (PCPA).The blank group were not treated,the model group were only bound without treatment,the acupuncture group were treated with acupuncture at Baihui,Shenmen,and Sanyinjiao,and the sham acupuncture group were treated with sham acupuncture at the same acupoints.The sleep latency and sleep time of rats before and after modeling and intervention were measured by pentobarbital sodium righting test.RT-PCR was used to detect expression of B-cell lymphoma-2 (Bcl-2),Bcl-2 associated X protein (Bax),and melatonin (MT) mRNA in the pineal gland,and immunohistochemistry were used to detect positive expression of Bcl-2 and Bax proteins in the pineal gland of rats.Results:After modeling,the sleep latency of insomnia rats were prolonged and the sleep time significantly shortened (all P<0.01).After treatment,the sleep latency of the acupuncture group were shortened and the sleep time prolonged (all P<0.01).Compared with the blank group,the expression of MT mRNA in the pineal gland of the model group were decreased,the expression of Bcl-2 protein and mRNA decreased,and the expression of Bax protein and mRNA increased (all P<0.05).Compared with the model group,the expression of MT mRNA in the pineal gland of the acupuncture group were significantly increased,the expression of Bcl-2 protein and mRNA increased,and the expression of Bax protein and mRNA were decreased (all P<0.05).There was no statistical difference in the sham acupuncture group ( P>0.05).Conclusion:Acupuncture may promote the secretion of MT in the pineal gland of insomnia rats by regulating the expression of apoptosis-related proteins,thereby shortening the sleep latency and prolonging the sleep time of PCPA-induced insomnia rats,and improving their sleep state.
