摘要:
目的:探讨通络活血汤调节Toll样受体4(TLR4)/核因子-κB(NF-κB)信号通路对输卵管阻塞性不孕症大鼠的干预作用。方法:将60只雌性大鼠随机分为正常组、输卵管阻塞组、低剂量通络活血汤组、高剂量通络活血汤组和高剂量通络活血汤+激活剂[25 μg/(kg·d) TLR4激动剂RS-09\]组,除正常组,其余组大鼠均构建输卵管阻塞性不孕症模型。酶联免疫吸附(ELISA)检测大鼠输卵管中白介素(IL)-1β、IL-6、IL-8、肿瘤坏死因子-α(TNF-α)表达;电子显微镜下观察输卵管超微结构;苏木素伊红(HE)染色观察输卵管病理变化;Western blot检测输卵管中TLR4/NF-κB通路蛋白表达;免疫组织化学(IHC)检测输卵管中p-NF-κB p65和p-IκBα蛋白表达水平。结果:与正常组比较,输卵管阻塞组大鼠输卵管损伤严重,IL-1β、IL-8、TNF-α和IL-6水平升高,TLR4、p-NF-κB p65和p-IκBα蛋白表达提高(P<0.05);与输卵管阻塞组比较,低剂量通络活血汤组和高剂量通络活血汤组大鼠输卵管损伤减轻,IL-1β、IL-8、TNF-α和IL-6水平降低,TLR4、p-NF-κB p65和p-IκBα蛋白表达降低(P<0.05),且高剂量通络活血汤组优于低剂量通络活血汤组(P<0.05);与高剂量通络活血汤组比较,高剂量通络活血汤+激活剂组大鼠输卵管损伤加重,IL-1β、IL-8、TNF-α和IL-6水平升高,TLR4、p-NF-κB p65和p-IκBα蛋白表达提高(P<0.05)。结论:通络活血汤通过抑制TLR4/NF-κB信号通路干预大鼠输卵管阻塞性不孕症。
Abstract:
Objective:To explore the intervention effect of Tongluo Huoxue decoction on fallopian tube obstructive infertility rats by regulating the Toll like receptor 4 (TLR4)/nuclear factor-κB (NF-κB) signaling pathway.Methods:Sixty female rats were randomly separated into normal group,fallopian tube obstruction group,low dose Tongluo Huoxue decoction group,high dose Tongluo Huoxue decoction group,and high dose Tongluo Huoxue decoction +activator \[25 μg/(kg·d) TLR4 activator RS-09\] group.Except for the normal group,rats in all other groups were used to construct models of fallopian tube obstructive infertility.Enzyme linked immunosorbent assay (ELISA) was performed to detect the expression of interleukin-1β,IL-6,IL-8,and tumor necrosis factor-α (TNF-α) in rat fallopian tubes.The ultrastructure of the fallopian tubes was observed under an electron microscope.Hematoxylin-eosin (HE) staining was performed to observe pathological changes in the fallopian tubes.Western blot was performed to detect the expression of TLR4/NF-κB pathway proteins in the fallopian tubes.Immunohistochemistry (IHC) was used to detect p-NF-κBp65 and p-IκBα protein expression levels in the fallopian tubes.Results:Compared with the normal group,the fallopian tube obstruction group had severe fallopian tube damage in rats,increased IL-1β,IL-8,TNF-α,and IL-6 levels,and TLR4,p-NF-κB p65,and p-IκBα protein expression (P<0.05).Compared with the fallopian tube obstruction group,the low dose Tongluo Huoxue decoction group and high dose Tongluo Huoxue decoction group showed reduced damage to the rat fallopian tubes,declined IL-1β,IL-8,TNF-α,and IL-6 levels,and TLR4,p-NF-κB p65,and p-IκBα protein expression (P<0.05),and the high dose Tongluo Huoxue decoction group was superior to the low dose Tongluo Huoxue decoction group (P<0.05).Compared with the high dose Tongluo Huoxue decoction group,the high dose Tongluo Huoxue decoction +activator group showed aggravated tubal injury in rats,increased IL-1β,IL-8,TNF-α,and IL-6 levels,and TLR4,p-NF-κB p65,and p-IκBα protein expression (P<0.05).Conclusion:Tongluo Huoxue decoction intervenes in fallopian tube obstructive infertility in rats by inhibiting the TLR4/NF-κB signaling pathway.
