摘要:
目的:探讨痔疮安膏的抗炎镇痛作用及其基于核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)炎症小体通路的作用机制。方法:采用巴豆油致大鼠肛周肿胀模型评价局部抗炎作用;通过二甲苯致小鼠耳肿胀、角叉菜胶致大鼠足肿胀及醋酸致小鼠扭体模型,分别评价全身抗炎与镇痛作用。采用冰醋酸诱导大鼠痔疮模型,HE染色与Masson染色观察肛周组织病理与纤维化;ELISA法检测血清炎症因子肿瘤坏死因子-α(TNF-α)、白介素(IL)-6、IL-1β、IL-18水平;Western blot检测肛周组织NLRP3、凋亡相关斑点样蛋白(ASC)、半胱氨酸天冬氨酸蛋白酶1(Caspase-1)及IL-1β蛋白表达。结果:痔疮安膏能剂量依赖性地抑制各类炎性肿胀及扭体反应(P<0.01或P<0.05)。痔疮安膏可显著减轻痔疮模型大鼠肛周组织炎细胞浸润、水肿及胶原沉积,降低血清促炎因子水平,并抑制NLRP3通路关键蛋白的表达(P<0.01或P<0.05)。结论:痔疮安膏具有明确的抗炎镇痛作用,其机制可能与抑制NLRP3炎症小体通路活化、减少促炎因子释放及改善组织纤维化有关。
Abstract:
Objective:To examine the anti-inflammatory and analgesic properties of Zhichuang’an ointment and its potential mechanism involving the NLRP3 inflammasome pathway.Methods:Anti-inflammatory activity was assessed using established models:croton oil-induced perianal edema in rats for local effects,and xylene-induced mouse ear edema along with carrageenan-induced rat paw edema for systemic effects.Analgesic efficacy was evaluated via the acetic acid-induced mouse writhing test.A rat hemorrhoidal model was generated by topical glacial acetic acid application.Perianal tissue histopathology and fibrosis were examined by HE and Masson staining.Serum concentrations of TNF-α,IL-6,IL-1β,and IL-18 were quantified by ELISA.Protein expression of NLRP3,ASC,Caspase-1,and IL-1β in tissues was analyzed by Western blot.Results:The ointment suppressed inflammatory swelling across all models and attenuated writhing responses in a dose-dependent manner (P<0.01 or P<0.05).In the hemorrhoid model,it markedly reduced inflammatory cell infiltration,tissue edema,and collagen deposition.It also decreased serum levels of pro-inflammatory cytokines and downregulated the expression of NLRP3 inflammasome pathway key proteins (P<0.01 or P<0.05).Conclusion:Zhichuang’an ointment demonstrates significant anti-inflammatory and analgesic effects,potentially mediated through the inhibition of NLRP3 inflammasome activation,leading to reduced release of pro-inflammatory cytokines and amelioration of tissue fibrosi.
