摘 要 目的:探讨复方红豆杉胶囊对Walker256移植性肝癌大鼠缺氧诱导因子(HIF1α)、血管内皮生长因子(VEGF)及增殖细胞核抗原(PCNA)表达的影响。方法:将50只SD大鼠随机分为空白组、模型组、低剂量组、中剂量组及高剂量组。对除空白组外其余组大鼠进行Walker256移植性肝癌造模,造模后第8 天对各组灌胃给予复方红豆杉胶囊,低剂量组、中剂量组及高剂量组给药剂量依次为0.93 g/kg,0.1875 g/kg及0.37 g/kg,连续21 d。末次给药24 h后,取各组大鼠血,检测其中肝功能指标\[天冬氨酸氨基转移酶(AST)、总胆红素(TBIL)、白蛋白(ALB)及丙氨酸氨基转移酶(ALT)\]。取各组大鼠肝癌病灶,对其进行病理组织学观测,并观察HIF1α、VEGF及PCNA免疫组化表达情况及相对表达量。结果:与模型组比,空白组、小剂量、中剂量和大剂量组大鼠血清ALB含量明显增高,而AST、TBIL及ALT含量则均明显降低(P<0.05)。与模型组比较,小剂量、中剂量和大剂量组大鼠肿瘤组织中肝癌细胞排列紊乱,胞核肿胀增大等现象明显减轻。与模型组比较,小剂量、中剂量和大剂量组大鼠肿瘤组织中HIF1α、VEGF及PCNA阳性表达情况均明显降低,且相对表达量均明显低于对照组(P<0.05)。结论:复方红豆杉胶囊对Walker256移植性肝癌大鼠HIF1α、VEGF及PCNA的表达具有较好的抑制作用。
Abstract:
Abstract Objective: To observe the effects of Fufang Hongdoushan capsule in the expression of HIF1α,VEGF and PCNA in Walker256 transplanted liver cancer mice. Methods: 50 SD mice were divided into control group,model group,low dose group,middle dose group and high dose group. Build the Walker256 transplanted liver cancer mice model in the groups except control group,after 8 days the model built,the Fufang Hongdoushan capsule was gavaged for 21 days with the dose of 0.93 g/kg,0.1875 g/kg and 0.37 g/kg in low dose group,middle dose group and high dose group,respectively. 24 hours after the last gavage,the levels of AST,TBIL,ALB and ALT were detected and the histopathologies of tumor issue were observed in the mice,while immunofluorescent staining of HIF1α,VEGF and PCNA were detected in the groups. Results: Compared with the model group,the levels of AST,TBIL,ALB and ALT were significantly promoted in low,middle and high dose groups (P<0.05). Pathological observations showed that the tumor cells in low,middle and high dose groups were significantly necrosis and apoptosis in the tumor tissue than which in the model group. Immunofluorescent staining shown,the positive situation and expression amounts of HIF1α,VEGF and PCNA were significantly lower in the low,middle and high dose groups than the model group (P<0.05). Conclusion: Fufang Hongdoushan capsule can promote the expression of HIF1α,VEGF and PCNA in Walker256 transplanted liver cancer mice.
