摘要:
摘 要 目的：观察益气化瘀散结方对胃癌SGC7901细胞增殖凋亡的影响，并通过益气化瘀散结方干预后凋亡相关蛋白Survivin、Bcl2的表达变化探讨其相关作用机制。方法： 体外培养SGC7901细胞，采用益气化瘀散结方含药血清干预，细胞分为空白血清组及5%、10%、20%益气化瘀散结方含药血清组，药物干预后，CCK8法检测细胞增殖情况，流式细胞术检测细胞凋亡，Westernblot、Realtime PCR 技术检测Survivin、Bcl2蛋白及mRNA的表达变化。结果：5%、10%、20%益气化瘀散结方含药血清组SGC7901细胞的增殖明显下降，与空白血清组比较，10%、20%浓度组差异显著（P＜0.05），48 h为实验最佳干预时间点；5%、10%、20% 益气化瘀散结方含药血清均能不同程度促进SGC7901细胞凋亡，尤以10%、20%含药血清组差异显著（P＜0.05）；10%、20%浓度益气化瘀散结方含药血清组Survivin、Bcl2蛋白及mRNA表达水平下降，与空白血清组比较差异显著(P＜0.05)。结论：益气化瘀散结方能抑制胃癌SGC7901细胞增殖、促进胃癌SGC7901细胞凋亡，其机制可能与降低凋亡抑制蛋白Survivin、Bcl2的表达相关。
Abstract:
Abstract Objective：To observe the effect of Yiqi Huayu Sanjie decoction on proliferation and apoptosis of cancer cell Line of SGC7901，and explore the mechanism by expression changes of Survivin and Bcl2 after intervention with Yiqi Huayu Sanjie decoction. Methods：SGC7901 cells were cultured in vitro and treated with drugcontaining serum of Yiqi Huayu Sanjie decoction，cells were devided into four groups：blank serum group，5%、10%、20% of drugcontaining serum of Yiqi Huayu Sanjie decoction groups，the proliferation rate was detected by CCK8，the cell apoptosis was detected by flow cytometry，the protein and mRNA expression of Survivin and Bcl2 were detected by Westernblot and Realtime PCR. Results：Compared with the blank serum group，each concentration of Yiqi Huayu Sanjie decoction could inhibit the proliferation of SGC7901 cells，in which 10% and 20% concentration significant especially (P＜0.05)，the most obvious time was 48 h;Compared with the blank serum group，each concentration of Yiqi Huayu Sanjie decoction could promote apoptosis of SGC7901 cells，especially the 10% and 20% concentration groups (P＜0.05)；Compared with the blank serum group，the expression of Survivin and Bcl2 protein and mRNA in serum of different concentration of Yiqi Huayu Sanjie decoction were significantly decreased(P＜0.05). Conclusion:Yiqi Huayu Sanjie decoction can inhibit the proliferation and promote apoptosis of gastric cancer SGC7901 cells，the mechanism of which may be related to the reduction of the expression of apoptosisinhibiting protein Survivin and Bcl2.
