摘 要 目的:研究中药复方扶脾化瘤饮诱导MFC胃癌小鼠细胞凋亡的作用及其对肿瘤组织中Bcl-2及Bax蛋白表达的影响,初步探讨其可能的抑瘤机制。方法:建立胃癌(MFC)小鼠动物模型,并将模型动物随机分为模型对照组、扶脾化瘤饮高、中、低剂量组共4组。扶脾化瘤饮高、中、低剂量组给药剂量分别为 6.0、3.0、1.5 g/kg ,模型组给予等体积纯净水。连续给药、给水 14 d 处死小鼠,分离肿瘤称重计算抑瘤率,部分采用流式细胞仪检测胃癌细胞凋亡率变化;部分石蜡包埋,肿瘤组织切片,免疫组织化学法检测 Bax、Bcl-2 蛋白表达。结果:经扶脾化瘤饮作用后,小鼠胃癌细胞凋亡率明显增加,且表现出良好的剂量关系。扶脾化瘤饮高、中剂量组显著上调肿瘤组织 Bax 蛋白表达,下调 Bcl-2 蛋白表达,与模型对照组比较,差异有统计学意义(P<0.05,P<0.01)。结论:扶脾化瘤饮可能通过下调Bcl-2表达,上调Bax表达,诱导肿瘤细胞凋亡,达到抗肿瘤治疗的目的。
Abstract:
Abstract Objective: To study the effect of Chinese herbal compound Fupi Hualiu decoction on apoptosis of MFC gastric cancer mice and its effect on the expression of Bcl-2 and Bax protein in tumor tissues, and to explore its possible anti-tumor mechanism. Methods: Animal models of gastric cancer (MFC) mice were established, and the model animals were randomly divided into model control group and Fupi Hualiu decoction high, medium and low dose groups. The doses of the high, medium and low doses of Fupi Hualiu decoction were 6.0g/kg, 3.0g/kg, 1.5g/kg, respectively, and the model group was given an equal volume of purified water. The mice were sacrificed by continuous administration and water supply for 14 days. The tumor inhibition rate was calculated by tumor weighing. The apoptosis rate of gastric cancer cells was detected by flow cytometry. Some paraffin-embedded, tumor tissue sections and immunohistochemical method were used to detect Bax and Bcl-2 protein expression. Results: After the treatment of Fupi Hualiu decoction , the apoptotic rate of gastric cancer cells increased significantly and showed a good dose relationship. The high and middle dose groups of Fupi Hualiu decoction significantly up-regulated the expression of Bax protein in tumor tissue and down-regulated the expression of Bcl-2 protein, which was statistically significant compared with the model control group (P<0.05, P<0.01). Conclusion: Fupi Hualiu decoction may induce the apoptosis of tumor cells by down-regulating the expression of Bcl-2 and up-regulating the expression of Bax, thus achieving the goal of anti-tumor therapy.
