摘要:
摘 要 目的:探讨圣愈汤联合放疗对人宫颈癌荷瘤小鼠脾脏DCs分泌细胞因子及CTLs对肿瘤细胞杀伤活性的影响。方法:建立小鼠皮下Hela宫颈癌细胞模型,随机分为对照组、圣愈汤组、放疗组、圣愈汤联合放疗组。治疗后7 d取各组小鼠脾脏制备细胞悬液,部分体外培养,ELISA法检测上清IFNγ的分泌情况;BD磁珠分选系统分离CD8+CTLs,与Hela宫颈癌细胞共培养,LDH法检测脾脏CD8+CTLs杀伤活性。结果:圣愈汤组、放疗组、圣愈汤联合放疗组脾脏DCs细胞活化数量较对照组升高,圣愈汤联合放疗组升高更明显(P<0.05)。圣愈汤组、放疗组、圣愈汤联合放疗组小鼠脾脏 IFNγ 分 泌 水 平 、脾脏 CD8+ CTLs 杀伤活性均较对照组升高,且以圣愈汤联合放疗组升高最为明显(P<0.05)。结论:圣愈汤可促进荷瘤小鼠脾DC的分化、成熟,增加INFγ分泌,诱导T淋巴细胞向CTLs的分化,增强CTLs杀伤活性,进而提高小鼠抗肿瘤免疫。
Abstract:
Abstract Objective:To explore the effect of Shengyu decoction combined with radiotherapy on cytokines secreted by spleen DCs and CTLs in mice with cervical carcinoma.Methods:A subcutaneous Hela cervical cancer cell model was established in mice, which was randomly divided into the control group, the shengyu decoction group, the radiotherapy group and the combination radiotherapy group.CD8+CTLs were isolated by BD magnetic bead sorting system and cocultured with Hela cervical cancer cells.Results:The activation amount of spleen DCs cells in the group of Shengyu decoction, radiotherapy group and the group of combination radiotherapy of Shengyu decoction was higher than that in the control group, and the increase was more obvious in the group of combination radiotherapy of shengyu decoction (P<0.05).Compared with the control group, the ifnfractionated water level and spleen CD8+CTLs killing activity of the spleen ifnfractionated rats of the Shengyu decoction group, the radiotherapy group and the combined radiotherapy group of the Shengyu decoction group all increased significantly (P<0.05).Conclusion:Shengyu decoction can promote the differentiation and maturation of spleen DC in tumorbearing mice, increase the secretion of infbeta, induce the differentiation of T lymphocytes into CTLs, enhance the killing activity of CTLs, and further improve the antitumor immunity of mice.
