摘 要 目的:对脑缺血再灌注损伤(CIRI)模型老年SD大鼠腹腔预先灌注麝香酮,检测实验后大鼠海马组织中S100β、NFκB p65活性值,了解麝香酮对脑组织核转录因子NFκB(NFκB)信号传导的影响。方法:采用健康SD老年大鼠60只。随机分为缺血再灌注组(CIRI),麝香酮组(SXT)和生理盐水对照组(CON)。腹腔灌注麝香酮,造缺血再灌注动物模型,处死大鼠后取海马区脑组织,采用凝胶电泳迁移分析法测定海马组织中S100β、NFκB p65活性值。实验分两次完成,每次完成30只大鼠实验。结果:CON、SXT与CIRI比较,S100β、NFκB p65都有明显下降,SXT与CON比较S100β的变化有明显差别,而NFκB p65的变化无明显差别。结论:麝香酮抗CIRI的作用可能与减少S100β蛋白释放,减少NFκB蛋白表达,减轻兴奋性氨基酸毒性有关。
Abstract:
Abstract 〖HT5”SS〗Objective:To cerebral ischemia reperfusion injury (CIRI) model old SD rat peritoneal perfusion musk ketone in advance,after the test S100β in rat hippocampus,NFκB p65 active value,understanding of musk ketone of brain tissue nuclear transcription factor NFκB (NFκB) the effects of signal transduction. Methods:Using 60 healthy SD senile rats. The rats were randomly divided into ischemia reperfusion group (CIRI),musk ketone group (SXT) and normal saline control group (CON). Abdominal cavity perfusion musk ketone,ischemia reperfusion animal models,to be put to death in the rat hippocampus after brain tissue,using the method of gel electrophoresis migration S100βin hippocampus,NFκB p65 active value. Experiment two complete,every 30 rats experiment.Results:Compared with CIRI,CON,SXT S100β,NFκB p65 had declined obviously,SXT compared with CON the change of S100βhave obvious difference,and NFκB p65 change had no obvious difference. Conclusion:The antiCIRI effect of musk ketone may be related to reducing the S100β protein release,reducing the expression of NFκB protein and reducing the toxicity of excitatory amino acid.
