摘要:
目的:本研究观察手十二井穴刺络放血对急性创伤性脑损伤(TBI)改善神经功能、减轻脑水肿有效的平台基础上,从抑制坏死性凋亡通路的角度出发,探索手十二井穴刺络放血在改善TBI中的活血化瘀作用及其机制。方法:采用电子控制性脑皮质撞击仪(eCCI),建立小鼠TBI模型,分别于造模后12、24、48、72 h观察手十二井穴刺络放血对TBI小鼠神经功能的影响,于造模后24 h通过HE染色观察手十二井穴刺络放血对TBI小鼠病理损伤的影响,于造模后12、24、48、72 h通过Western blot检测,观察手十二井穴刺络放血对TBI小鼠坏死性凋亡相关蛋白的影响。结果:各组小鼠创伤前 mNSS 评分比较,差异无统计学意义(P>0.05),具有可比性; 与假手术组相比,模型组的mNSS评分在创伤后12、24、48、72 h均显著升高(P<0.01),井穴放血组的mNSS 评分在创伤后12、24、48、72 h均显著低于模型组(P<0.01),Western blot检测发现,与假手术组相比,模型组RIP-1、RIP-3、MLKL蛋白表达在颅脑创伤后6、24、48、72 h均显著升高(P<0.01),与模型组相比,井穴放血组RIP-1、RIP-3、MLKL蛋白表达在颅脑创伤后6 、24 、48、72 h均显著降低(P<0.01)。结论:手十二井穴刺络放血可通过抑制RIP-1、RIP-3、MLKL蛋白表达,缓解炎症反应,发挥神经保护作用,改善神经功能缺损等相关症状。
Abstract:
Objective:This study is based on the effective platform of blood-letting puncture of hand-twelve-well points on TBI to improve neurological function and reduce brain edema,and to explore the “promoting blood circulation and removing blood stasis” of hand-twelve-well acupuncture in improving TBI and its mechanism from the point of view of inhibiting necrotizing apoptosis pathway.Methods:The mouse TBI model was established by electronically controlled cortical percussion instrument.The effects of puncture and bloodletting on the neurological function of TBI mice were observed at 12,24,48 and 72 h after modeling.The pathological injury of TBI mice was observed by HE staining at 24 h after modeling.The pathological injury of TBI mice was detected by Westernblot at 12,24,48 and 72 h after modeling.To observe the effect of blood-letting puncture at hand twelve well points on necrotizing apoptosis-related proteins in TBI mice.Results:There was no significant difference in mNSS score before trauma in each group.Compared with the sham operation group,the mNSS score of the model group increased significantly at 12,24,48 and 72 h after trauma(P<0.01).The mNSS score of the well acupoint bloodletting group was significantly lower than that of the model group at 12,24,48 and 72 h after trauma(P<0.01).Compared with the sham operation group,the mNSS score of the well acupoint bloodletting group was significantly lower than that of the model group(P<0.01).The protein expression of RIP-1,RIP-3 and MLKL in the model group increased significantly at 6,24,48 and 72 h after craniocerebral trauma.Compared with the model group,the protein expression of RIP-1,RIP-3 and MLKL in the well acupoint bloodletting group decreased significantly at 6,24,48 and 72 h after craniocerebral trauma.Conclusion:Blood-letting puncture at hand twelve well points can alleviate inflammatory reaction,exert neuroprotective effect and improve related symptoms such as nerve function defect by inhibiting the expression of RIP1,RIP3 and MLKL protein.
