摘要:
目的:对柴枳夏及汤作用于胆汁反流性胃炎(BRG)大鼠模型胃肠动力的影响进行研究,并对其作用机制进行探讨。方法:选用Wistar大鼠50只,用随机数字表法并按照大鼠体重分成五组,分别为西药组(达喜和莫沙必利)、柴枳夏及汤低剂量组(生药量12.77 g/kg)、柴枳夏及汤高剂量组(生药量25.54 g/kg)、模型组以及空白组,除空白组外,灌胃自制反流液制备胆汁反流性胃炎大鼠模型。观察空白组及模型组大鼠一般状态,体重,肉眼观察其胃黏膜变化; 检测胃排空率及小肠推进率; 通过ELISA法检测BRG大鼠血清及血浆中胃动素(MTL)、血管活性肠肽(VIP)及生长抑素(SS)含量变化。结果:与空白组比较,模型组大鼠出现排便稀糊、便中含红黄色黏液、反应迟钝、体重增长缓慢(P<0.05)等特点; 肉眼可见胃窦部点片状糜烂,有黄绿色胆汁及较多黄色黏液; 模型组大鼠胃排空率和小肠推进率均显著降低(P<0.01),MTL含量明显下降(P<0.01),VIP及SS含量均显著升高(P<0.01)。与模型组比较,柴枳夏及汤高剂量组能够显著提高胆汁反流性胃炎大鼠MTL含量(P<0.01),能够显著降低其VIP及SS含量(P<0.01),同时显著提升胃排空率及小肠推进率(P<0.01或P<0.05)。结论:柴枳夏及汤具有良好的促胃肠动力作用,其机制可能与调节MTL、VIP、SS等胃肠激素的水平有关。
Abstract:
Objective:To investigate the effect and mechanism of Chaizhi Xiaji decoction on experimental bile reflux gastritis(BRG)rats.Methods:Five groups were randomly set up according to the weight of Wistar rats:blank group,model group,Chaizhi Xiaji decoction of low and high dose groups,and western medicine group(Hydrotalcite tablets and Mosapride Citrate tablets).The experimental bile reflux gastritis rats were induced by given self-made reflux liquid orally except rats in blank group.The general state and body weight of rats in blank group and model group were observed.The changes of gastric antrum mucosa were observed by naked eye.The gastric motility and the small intestine propulsion rate were measured and compared with each group.ELISA method was used for detection of MTL,VIP and SS content.Results: Compared with blank group,rats in model group showed thin stool containing red yellow mucus,slow reaction,and body weight decreased significantly(P<0.05).There were patchy erosions of the gastric antrum with yellow green bile and more yellow mucus of model group in the naked eye.Gastric emptying and intestinal propulsion of model group were slowed down significantly(P<0.01).The content of MTL in serum and plasma of model group decreased significantly(P<0.01)and the content of VIP and SS significantly increased(P<0.01).Compared with model group,Chaizhi Xiaji decoction of high dose group significantly increased the content of MTL in serum and plasma of BRG rats(P<0.01).And the content of SS in plasma and serum were decreased under the influence of Chaizhixiaji decoction of high dose group,as well as the content of VIP(P<0.01).Moreover,the rate of gastric emptying and intestinal propulsion were increased significantly(P<0.01 or P<0.05).Conclusion: Chaizhi Xiaji decoction has obvious effect on promoting gastrointestinal dynamics,and its mechanism may be related with the regulation of MTL,VIP and SS contents.
