摘要:
目的:探讨加味小陷胸汤基于核因子κB(NF-κB)信号通路对非小细胞肺癌荷瘤小鼠的影响。方法:选取SPF级健康雄性C57BL/6小鼠75只,分为对照组15只和造模组60只。造模组采用人非小细胞肺癌A549细胞株右腋下接种建立非小细胞肺癌荷瘤模型,对照组给予等量生理盐水右腋下皮下注射。造模成功后,将造模组小鼠随机分为模型组、加味小陷胸汤低、高浓度组及顺铂组,各15只。模型组和对照组分别予以无菌生理盐水5 ml/kg,腹腔注射,1次/d +蒸馏水5 ml/kg,灌胃,1次/d; 加味小陷胸汤低、高浓度组分别予以无菌生理盐水5 ml/kg,腹腔注射,1次/d +8 mg/ml、16 mg/ml加味小陷胸汤药液5 ml/kg,灌胃,1次/d; 顺铂组予以0.4 mg/ml顺铂溶液5 ml/kg,腹腔注射,1次/d +蒸馏水5 ml/kg,灌胃,1次/d。所有小鼠均连续给药14 d。对小鼠肿瘤体积、瘤重和生长抑制率、肿瘤组织细胞凋亡率、肿瘤组织含半胱氨酸的天冬氨酸蛋白水解酶-3(Caspase-3)、NF-κB、B淋巴细胞瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)表达水平进行检测。结果:治疗后,与模型组比较,各组小鼠肿瘤体积、瘤重较低,肿瘤组织NF-κB、Bcl-2表达水平较低,且加味小陷胸汤低浓度组>加味小陷胸汤高浓度组和顺铂组,差异有统计学意义(P<0.05)。与模型组比较,各组小鼠肿瘤生长抑制率较高,肿瘤组织细胞凋亡率较高,肿瘤组织Bax、Caspase-3表达水平较高,且加味小陷胸汤低浓度组<加味小陷胸汤高浓度组和顺铂组,差异有统计学意义(P<0.05)。结论:加味小陷胸汤能剂量依赖性抑制非小细胞肺癌荷瘤小鼠肿瘤生长,促进肿瘤细胞凋亡,其作用可能与抑制NF-κB信号通路,调节凋亡相关蛋白Bcl-2、Bax、Caspase-3表达有关。
Abstract:
Objective:To analysis the effect of Jiawei Xiaoxianxiong decoction on non-small cell lung cancer mice based on nuclear factor-κB(NF-κB)signaling pathway.Methods:75 SPF healthy male C57BL/6 mice were selected,which were divided into control group with 15 mice and model building group with 60 mice.Non-small cell lung cancer models were established by right axillary inoculation of human non-small cell lung cancer A549 cell line in model building group,and mice in the control group were given the same amount of saline by subcutaneous injection under right axilla.After the establishment of the model,mice in model building group were randomly divided into model group,Jiawei Xiaoxianxiong decoction low-concentration group,Jiawei Xiaoxianxiong decoction high-concentration group and cisplatin group,each with 15 mice.The model group and control group were given aseptic saline 5 ml/kg,ip,qd+distilled water 5 ml/kg,ig,qd; Jiawei Xiaoxianxiong decoction low-concentration group,Jiawei Xiaoxianxiong decoction high-concentration group were given aseptic saline 5 ml/kg,ip,qd+8 mg/ml,16 mg/ml Xiaoxianxiong decoction suspension 5 ml/kg,ip,qd; cisplatin group was given 0.4 mg/ml cisplatin solution 5 ml/kg,ip,qd + distilled water 5 ml/kg,ig,qd.All mice were given 14 d of continuous administration.The tumor volume,tumor weight and growth inhibition rates,apoptosis rates of tumor tissue,expression levels of cysteine-containing aspartate proteolytic enzyme-3(caspase-3),NF-κB,Bcl-2 and Bcl-2 related X protein(Bax)in tumor tissues were detected.Results:After treatment,compared with model control group,the tumor volume and tumor weight were lower,expression levels of NF-κB,Bcl-2 in tumor tissues were lower in each group,and Jiawei Xiaoxianxiong decoction low-concentration group > Jiawei Xiaoxianxiong decoction high-concentration group and cisplatin group,the difference was statistically significant(P<0.05).Compared with model control group,the inhibition rates of tumor growth were higher,the apoptotic rates of tumor tissue were higher,and the expression levels of Bax and Caspase-3 in tumor tissue were higher in each group,and Jiawei Xiaoxianxiong decoction low-concentration group < Jiawei Xiaoxianxiong decoction high-concentration group and cisplatin group,the difference was statistically significant(P<0.05).Conclusions:Jiawei Xiaoxianxiong decoction can inhibit the growth of non-small cell lung cancer-bearing mice in a dose-dependent manner,promote apoptosis of cancer cells,the effect may be related to inhibiting the signal pathway of NF-κB,and regulating the expressions of apoptosis-related proteins Bcl-2,Bax and Caspase-3.
