摘要:
目的:探讨止麻消痰活血汤对心肌梗死无复流大鼠的治疗效果。方法:选择健康雄性大鼠60只,随机分为对照组、模型组和止麻消痰活血汤组,各20只。除对照组外,其他两组通过冠状动脉结扎方法建立心肌梗死无复流模型,自造模之日起,对照组和模型组给予生理盐水灌胃,止麻消痰活血汤组给予止麻消痰活血汤干预。造模成功4周后,观察各组心功能情况; ELISA法检测各组血清中IL-6、CRP、TNF-α水平情况,采用Western blot法检测心肌组织中NF-κB p65蛋白及其磷酸化水平情况。结果: 实验4周后,与对照组相比,模型组和止麻消痰活血汤组的LVESd及LVEDd明显增加,LVEF及LVFS明显降低,且模型组比对照组更明显(P<0.05); 实验4周后,模型组和止麻消痰活血汤组IL-6、CRP、TNF-α水平高于对照组,且模型组高于止麻消痰活血汤组(P<0.05)。NF-κB p65及phospho-NF-κB p65蛋白水平表达情况:对照组<止麻消痰活血汤组<模型组(P<0.05)。 结论:止麻消痰活血汤可通过抑制心肌梗死无复流大鼠NF-κB依赖的机制,抑制心脏重塑,改善其心功能,降低心肌炎症因子水平,从而改善无复流现象。
Abstract:
Objective:To investigate the effect of Zhima Xiaotan Huoxue decoction on NF-κB p65 protein and its phosphorylation level in rats with myocardial infarction and no reflow.Methods:60 healthy male rats were randomly divided into control group,model group and Zhima Xiaotan Huoxue decoction group with 20 rats each.In addition to the control group,the other two groups established no reflow model of myocardial infarction by coronary artery ligation.From the day of modeling,the normal group and model group were given normal saline,Zhima Xiaotan Huoxue decoction group were given Zhima Xiaotan Huoxue decoction intervention.Four weeks after the model was established successfully,the cardiac function of each group was observed; the levels of IL-6,CRP and TNF-α in serum of each group were detected by ELISA,and the levels of NF-κB p65 protein and its phosphorylation in myocardial tissue were detected by Western blot.Results:After 4 weeks,compared with the control group,the LVESD and lved of model group and Zhima Xiaotan Huoxue decoction group increased significantly,the LVEF and LVFS of model group decreased significantly,and the level of IL-6,CRP and TNF-α of model group and Zhima Xiaotan Huoxue decoction group were higher than that of control group(all P<0.05).The expression of NF-κB p65 and phospho-NF-κB p65 protein in the control group was lower than that in the model group(all P<0.05).Conclusion:Zhima Xiaotan Huoxue decoction can improve the no reflow phenomenon by inhibiting the mechanism of NF-κB dependence,inhibiting the cardiac remodeling,improving the cardiac function and reducing the level of myocarditis factor.
