摘要:
目的:研究防己黄芪汤对人三阴性乳腺癌细胞MB-MDA-231增殖、凋亡以及侵袭力的影响。方法:MTT法检测防己黄芪汤对人三阴性乳腺癌细胞MB-MDA-231的抑制作用; 倒置显微镜下观察细胞形态变化。将实验组分成4、8、16 mg/ml的不同浓度组,AnnexinV-FITC /PI双染检测MB-MDA-231细胞凋亡; Transwell法测定细胞体外迁移能力。结果: 防己黄芪汤可显著抑制MB-MDA-231细胞生长,在4、8、16、32、64 mg/ml浓度范围内呈浓度依赖性。防己黄芪汤能明显改变细胞形态,倒置显微镜下仅可见很少的贴壁细胞,多散在排列,部分细胞体积改变,胞浆内出现空泡变性,细胞结构不完整,裂解成碎片。防己黄芪汤作用24 h能够提高MB-MDA-231细胞的总凋亡率、早期凋亡率和晚期凋亡率,其中提高早期凋亡率更为明显。抑制MB-MDA-231细胞体外迁移的能力,高浓度组效果更佳,迁移细胞数明显减少。结论:防己黄芪汤能够抑制人三阴性乳腺癌细胞MB-MDA-231的增殖、迁移,并且促进其凋亡。
Abstract:
Objective:To research the effects of Fangji Huangqi decoction to triple negative breast cancer MB-MDA-231 in vitro.Methods:MTT was used to detect the inhibitory effect of Fangji Huangqi decoction on human triple-negative breast cancer cell MB-MDA-231.Observing the cell morphological changes by inverted microscope.The test group were divided into 4,8,16 mg/ml,low,medium,and high concentration groups.Annexin V-FITC/PI double staining was used to detect the apoptosis of MB-MDA-231 cells.The Transwell was used to determine the cell migration ability in vitro.Results:Fangji Huangqi decoction can significantly inhibit the growth of MB-MDA-231 cells in a concentration-dependent range of 4,8,16,32 and 64 mg/ml.Fangji Huangqi decoction can significantly change the morphology of cells.Only a few adherent cells can be seen under an inverted microscope.They are mostly scattered and arranged,some of the cells have changed in volume,vacuolar degeneration appears in the cytoplasm,and the cell structure is incomplete and broken into fragments.Fangji Huangqi decoction can increase the total apoptosis rate,early apoptosis rate and late apoptosis rate of MB-MDA-231 cells after 24 hours,and the improvement of early apoptosis rate is more obvious.The high concentration group has a better effect on inhibiting the ability of MB-MDA-231 cells to migrate in vitro,and the number of migrated cells is significantly reduced.Conclusion:Fangji Huangqi decoction can significantly inhibit MB-MDA-231,cause MB-MDA-231 apoptosis and nhibit the invasiveness of MB-MDA-231 in vitro.
