摘要:
目的:探讨藿苓益智方对阿尔茨海默病线粒体通路神经细胞凋亡的调控作用。方法:采用Aβ的活性片段Aβ25-35诱导PC12细胞凋亡建立阿尔茨海默病的细胞模型。本实验设置六个组,即对照组、Aβ25-35组、Aβ25-35+盐酸多奈哌齐组、Aβ25-35+藿苓益智方提取物(50、100、200 μg/ml)组。利用流式细胞仪、荧光酶标仪和透射电镜等技术,检测霍苓益智方提取物对阿尔茨海默病细胞模型中细胞存活率、神经细胞凋亡率、线粒体超微结构的变化。结果:藿苓益智方提取物50 μg/ml组的细胞存活率与Aβ25-35组相比有一定的增高(P<0.05); 盐酸多奈哌齐组和藿苓益智方提取物(100、200 μg/ml)组与Aβ25-35组相比,细胞存活率显著升高(均P<0.01),但藿苓益智方提取物200 μg/ml组升高的更明显; 藿苓益智方低、中、高剂量+A25-35组细胞凋亡率则逐步下降,其中中、高剂量组与Aβ25-35组相较差异均具有统计学意义(均P<0.01); 藿苓益智方低、中、高剂量+Aβ25-35组对细胞核的损害程度上,经治疗以后逐步缓解,且呈剂量依赖性趋势。结论:藿苓益智方对阿尔茨海默病的治疗中可对Aβ诱导的PC12细胞凋亡具有明显的抑制作用,改善细胞存活率,减少线粒体细胞凋亡。
Abstract:
Objective:To investigate the regulatory effect of Huoling Yizhi recipe on apoptosis of neurons in mitochondrial pathway.Methods:The apoptosis of PC12 cells was induced by Aβ25-35,the active fragment of Aβ,to establish the cell model of Alzheimer disease.In this experiment,six groups were set up:control group,Aβ25-35 group,Aβ25-35+donepezil hydrochloride group,Aβ25-35+Huoling Yizhi extract(50,100,200 μg/ml)group.The changes of cell survival rate,neuron apoptosis rate and mitochondrial ultrastructure in Alzheimer disease cell model were detected by flow cytometry,fluorescent enzyme labeling and transmission electron microscopy.Results:Compared with Aβ25-35 group,the cell survival rate of the 50 μg/ml group was higher(P<0.05); Compared with the Aβ25-35 group,the cell survival rate of the donepezil group and the 100,200 μg/ml group was significantly higher(all P<0.01),but the 200 μg/ml group was more significantly higher; the low,medium and high dose+Aβ25-35 group the apoptotic rate decreased gradually,and there was a significant difference between the middle and high dose groups and Aβ25-35 group(all P<0.01); The degree of nuclear damage in the low,middle and high dose+Aβ25-35 group of Huoling Yizhi ricipe was gradually relieved after treatment,and showed a dose-dependent trend.Conclusion:In the treatment of Alzheimer disease,Huoling Yizhi recipe can obviously inhibit the apoptosis of PC12 cells induced by Aβ,improve the cell survival rate and reduce the apoptosis of mitochondrial cells.
