摘要:
目的:评估益智醒脑方对阿尔茨海默病大鼠学习记忆能力及对白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平的影响。方法:将90只老年Wistar大鼠随机分为六组含高剂量益智醒脑方组(高剂量组)、中剂量益智醒脑方组(中剂量组)、低剂量益智醒脑方组(低剂量组)、正常组、模型组及西药组,每组15只,除了正常组其余各组均进行Aβ1-42造模制备成阿尔茨海默病大鼠。观察各组大鼠学习记忆力、海马APP、VPS35、SorLA的表达及海马炎症因子IL-1β、IL-6、TNF-α水平。结果:高剂量组及正常组穿越平台次数高于模型组、西药组、中剂量组及低剂量组,差异均有统计学意义(P<0.05); 高剂量组逃避潜伏期低于模型组、正常组、西药组、中剂量组及低剂量组,差异均有统计学意义(P<0.05)。正常组APP、VPS35、SorLA的表达均高于高剂量组、模型组、西药组、中剂量组及低剂量组,差异均有统计学意义(P<0.05)。正常组IL-1β、IL-6、TNF-α水平均低于高剂量组、模型组、西药组、中剂量组及低剂量组,差异均有统计学意义(P<0.05)。结论:益智醒脑方能够改善阿尔茨海默病大鼠学习记忆能力,降低IL-1β、IL-6、TNF-α水平。
Abstract:
Objective:To evaluate the effects of Yizhi Xingnao recipe on the learning and memory abilities of Alzheimer's disease rats and the levels of Interleukin-1β(IL-1β),Interleukin-6(IL-6)and Tumor necrosis factor-α(TNF-α).Methods:90 aged Wistar rats were randomly divided into 6 groups including high-dose Yizhi Xingnao recipe group(high-dose group),medium-dose Yizhi Xingnao recipe group(medium-dose group),and low-dose Yizhi Xingnao recipe group(low-dose group),normal group,model group and western medicine group,15 rats in each group,except for the normal group,all other groups were modeled with Aβ1-42 to prepare Alzheimer's disease rats.The learning and memory ability,the expression of hippocampal APP,VPS35,SorLA and the levels of hippocampal inflammatory factors IL-1β,IL-6,and TNF-α were observed in each group of rats.Results:The frequency of crossing the platform in high-dose group and normal group was higher than that in model group,western medicine group,medium-dose group and low-dose group,and the difference was statistically significant(P<0.05).The escape latency of high-dose group was lower than those in model group,normal group,western medicine group,medium-dose group and low-dose group,and the difference was statistically significant(P<0.05).The expressions of APP,VPS35 and SorLA in normal group were higher than those in high-dose group,model group,western medicine group,medium-dose group and low-dose group,with statistical significance(P<0.05).The levels of IL-1β,IL-6 and TNF-α in normal group were lower than those in high-dose group,model group,western medicine group,medium-dose group and low-dose group,with statistical significance(P<0.05).Conclusion:Yizhi Xingnao recipe can improve the learning and memory ability of Alzheimer's disease rats,and reduce the levels of IL-1β,IL-6 and TNF-α.
