摘要:
目的:探讨丹参、柴胡、五味子、灵芝四种中药不同药性与药理学作用机制的关系。方法:采用20%四氯化碳(CCl4)香油溶液多次注射,饮用10%乙醇溶液并以高脂低蛋白饲料饲养小鼠制备肝郁脾虚模型,分别给予不同药性药物治疗10 d,酶法测定各实验组小鼠血清和肝匀浆中谷丙转氨酶(ALT)、乳酸脱氢酶(ALP)、胆碱酯酶(CHE)、谷胱甘肽(GSH); 体外培养肝星状细胞系(HSC-T6)并采用TGF-β1诱导其活化,Western blotting检测各实验组α-SMA、Ras、CollagenⅠ、ERK、p-ERK、TβRⅠ、TβRⅡ、Smad2/3蛋白的表达,观察药性性能不同的中药对HSC-T6表达Ras/ERK、TGF-β1/Smad信号通路蛋白的影响。结果:药性苦、寒柴胡,药性苦、微寒丹参处理肝郁脾虚模型小鼠,其生物学效应指标与中药药性论大致相似。药性酸、甘、温五味子处理肝郁脾虚模型小鼠,其生物学效应指标与中药酸、甘、温药性不尽一致,药性甘、平灵芝与前三者药性不同,其生物学效应指标显著差异。不同药性的药物处理HSC-T6靶细胞内Ras/ERK与TGF-β1/Smad信号通路蛋白表达存在着调节差异性。结论:依据中药药性的描述,通过研究影响机体生理变化和靶细胞内信号蛋白表达的意义,探讨不同药性的药理学作用机制,辨证地为广义中药药性赋予了药理学科学内涵。
Abstract:
Objective:To explore the relationship between the different medicinal properties of four traditional Chinese medicines(TCM)and their pharmacological mechanism with Salvia miltiorrhiza,Bupleurum,Schisandrae and Ganoderma lucidum.Methods:The model of liver depression and spleen deficie was prepared by injecting carbon tetrachloride(CCl4)and drinking 10% ethanol solution and feeding the mice with high-fat and low-protein diet.The mice were treated with different drugs for 10 days,respectively.ALT,ALP and CHE in serum and liver homogenate of the mice in each experimental group were determined by enzymatic method.HSC-T6 cell line was cultured in vitro and activated induced by the TGF-β1,Western blotting was used to detect the expressions of α-SMA,Ras,Collagen Ⅰ,ERK,p-ERK,TβRⅠ,TβRⅡ,Smad2/3 in each experimental group.To observe the effect of different properties of Chinese medicines on the expressions of Ras/ERK and TGF-β1/Smad signaling pathway proteins in HSC-T6 cell line.Results:The model mouse of liver depression and spleen deficie was treated by bitter,cold bupleurum and bitter,slight cold salvia miltiorrhiza,the biological effect indexs were roughly similar to the properties of TCM.The model mouse was treated by Sour,sweet,benign schisandrae,the biological effect indexs not consistent with the properties of TCM.The biological effect indexes of sweet,soft ganoderma lucidum were significantly different.Hsc-t6 cells treated with different drugs showed regulatory differences in expression of Ras/ERK and TGF- β1/Smad signaling pathway proteins.Conclusion:According to the description of the medicinal properties of TCM by investigating the significance of influencing the physiological changes of the body and the expression of signal proteins in target cells.The pharmacology mechanism of different medicinal properties was investigated,dialectically,pharmacology is endowed with scientific connotations for the broad sense of medicinal property of TCM.
