摘要:
目的:探讨益气活血方对乳腺癌MCF-7细胞系增殖侵袭能力及microRNA-100(miR-100)表达水平的影响。方法:取对数生长期的MCF-7乳腺癌细胞,将益气活血方以培养液稀释调整为20、40、80 mg/L培养细胞,设置高、中和低浓度组,并以不含药物的相同培养液培养细胞为对照组。分别采用qRT-PCR检测miR-100的表达水平; MTT法检测细胞增殖; Transwell小室法检测细胞侵袭; 并采用Western blot法检测细胞增殖相关蛋白特异性周期蛋白D1(Cyclin D1)和侵袭相关蛋白基质金属蛋白酶-9(MMP-9)的表达水平。结果:与对照组相比,实验组miR-100表达水平升高; 且与较低浓度组相比,较高浓度组miR-100表达水平亦呈上升趋势(均P<0.05)。与对照组相比,实验组细胞增殖抑制率和侵袭抑制率明显升高,且与较低浓度组相比,较高浓度组各抑制率均亦呈上升趋势(均P<0.05)。同时,与对照组相比,实验组Cyclin D1和MMP-9蛋白表达水平明显降低,且与较低浓度组相比,较高浓度组Cyclin D1和MMP-9蛋白表达水平亦呈下降趋势(均P<0.05)。结论:益气活血方可抑制乳腺癌细胞的增殖和侵袭。其机制可能与益气活血方可提高miR-100表达水平、降低Cyclin D1和MMP-9表达水平相关。
Abstract:
Objective:To investigate the effect of Yiqi Huoxue recipe on the proliferation and invasion ability of breast cancer MCF-7 cell line and its impact on the expression level of microRNA-100(miR-100).Methods:MCF-7 breast cancer cell lines in logarithmic growth phase treated by culture medium diluted with Yiqi Huoxue recipe at 20,40 and 80 mg/L,with the establishment of three groups of high,medium and low concentration experimental groups.Cells treated with the same culture medium without drugs were used as the control.qRT-PCR was used to detect the expression of miR-100; MTT method was used to detect cell proliferation; Transwell chamber method to detect cell invasion; and Western blot to detect the expression of cell proliferation related protein specific cyclin D1(cyclin D1)and invasion related protein matrix metalloproteinase-9(MMP-9).Results:Compared with the control group,the expression level of miR-10 increased in the experimental group,and the expression level in the higher concentration group also showed a reduced trend compared with the lower concentration group(all P<0.05).Compared with the control group,the inhibition rates of cell proliferation and invasion in the experimental group were significantly increased,which also showed an upward trend in the higher concentration group when compared with the lower concentration group(all P<0.05).In addition,in relative to the control group,the protein expression levels of Cyclin D1 and MMP-9 in the experimental group were significantly lower,which also decreased in the higher concentration group than those in the lower concentration group(all P<0.05).Conclusion:Yiqi Huoxue recipe can suppress the proliferation and invasion of breast cancer cells.Its mechanism of action may be related to its impact on the increase of the expression of miR-100,and the decrease of the expression of Cyclin D1 and MMP-9.
