摘要:
目的:基于网络药理学方法探讨茵陈蒿汤治疗高脂血症的作用机制。方法:使用TCMSP数据库筛选出茵陈蒿汤主要活性成分及作用靶点; 使用Gencards、Drugbank、OMIM数据库筛选高脂血症靶基因,利用韦恩图取交集得到潜在作用靶点; 利用String平台分析靶点蛋白的交互关系; 通过Metascape平台进行GO富集和KEGG富集分析其作用机理; 并利用Cytoscape构建“活性成分-靶点-通路”可视化网络图。结果:共筛选茵陈蒿汤17个有效成分,57个调治高脂血症潜在靶点; PPI网络中Degree值前3名的核心靶点分别为IL-6、AKT1、TNF; GO与KEGG富集主要通过细胞对活性氧代谢、脂质反应、对脂多糖反应等过程及AGE-RAGE、流体剪切应力与动脉粥样硬化、HIF-1等信号通路发挥其生物效应,体现了茵陈蒿汤多成分、多靶点、多通路调治高脂血症的特点。结论:茵陈蒿汤通过抑制脂肪生成、代谢,促进胆固醇排泄,抗氧化,调节炎症反应血管重塑等发挥调节血脂的作用。
