摘要:
目的:探讨不同剂量六味地黄丸对胰岛素抵抗小鼠糖代谢及体重的影响。方法:应用不同剂量六味地黄丸干预高脂饮食诱导的胰岛素抵抗小鼠。药物干预6周后检测小鼠糖耐量。干预8周后检测空腹血糖,留取血清检测空腹胰岛素,计算胰岛素抵抗指数(HOMA-IR); 检测未羧化骨钙素(ucOC)、羧化骨钙素(cOC),计算两者之间的比值; 计算药物干预前后小鼠的体重增幅。对HOMA-IR、骨钙素、体重增幅两两之间做相关性分析。结果:与模型组比较,六味地黄丸各剂量组均能够改善胰岛素抵抗小鼠的糖耐量,尤以六味地黄丸小剂量组的作用显著。与模型组比较,六味地黄丸小剂量组和六味地黄丸大剂量组可改善胰岛素抵抗小鼠的HOMA-IR(均P<0.01),且六味地黄丸小剂量组具有更好的改善作用。与模型组比较,六味地黄丸小剂量组治疗前后体重增幅最小,差异有统计学意义(P<0.05)。六味地黄丸中、大剂量组体重增幅与模型组比较,差异无统计学意义(均P>0.05)。体重增幅与HOMA-IR呈正相关(r=0.711,P<0.01); 体重增幅与ucOC/cOC呈负相关(r=-0.387,P<0.01),但与ucOC无相关性(r=-0.141,P>0.05); HOMA-IR与ucOC/cOC呈负相关(r=-0.358,P<0.05),但与ucOC无相关性(r=-0.156,P>0.05)。结论:六味地黄丸小剂量组可更好改善胰岛素抵抗小鼠糖耐量和胰岛素抵抗,对体重增幅的影响最小,其作用可能与ucOC/cOC有关。
Abstract:
Objective:To investigate effects of different doses of Liuwei Dihuang pill on glucose metabolism and body weight in insulin resistant mice.Methods:Different doses of Liuwei Dihuang pill were used to intervene in high-fat diet-induced insulin resistance mice.Glucose tolerance was measured after 6 weeks of drug intervention.Fasting blood glucose was measured after 8 weeks of intervention and serum taken to detect insulin,uncarboxylated osteocalcin(unOC)and carboxylated osteocalcin(cOC).Insulin resistance index(HOMA-IR)and the ratio of unOC to cOC was calculated(ucOC/cOC),weight gain before and after drug intervention was recorded.Correlation analysis was made between the following indicators such as HOMA-IR,osteocalcin and weight gain.Results:Compared with the model group,each dose of Liuwei Dihuang pill group can improve the glucose tolerance of insulin resistant mice,especially in the low dose group.Compared with the model group,both the low-dose group and high-dose group of Liuwei Dihuang pill can improve HOMA-IR in insulin-resistant mice(all P<0.01).The low-dose group may have better effect.The low-dose group of Liuwei Dihuang pill group had the smallest weight gain,difference statistically significant compared with the model group(P<0.05).The other two groups of Liuwei Dihuang pill group had no statistically significant difference compared with the model group(all P>0.05).Correlation analysis showed that weight gain was significantly positively correlated with HOMA-IR(r=0.711,P<0.01).Weight gain was significantly negatively correlated with ucOC/cOC(r=-0.387,P<0.01),but no significant correlation with ucOC(r=-0.141,P>0.05).HOMA-IR was significantly negatively correlated with ucOC/cOC(r=-0.358,P<0.05),but no significant correlation with ucOC(r=-0.156,P>0.05).Conclusion:The low dose group of Liuwei Dihuang pill could better improve the glucose tolerance and insulin resistance of insulin-resistant mice with the least effect on body weight.Its effect may be related to ucOC/cOC.
