摘要:
目的:研究益心附葶饮对腹主动脉缩窄致心力衰竭大鼠心肌纤维化的改善作用及机制。方法:采用腹主动脉缩窄法构建心衰大鼠模型,将造模成功的30只大鼠随机分为模型组和中药组,另设15只穿线不结扎的假手术组。中药组给予益心附葶饮灌胃3.5 g/(kg·d),假手术组和模型组给予等体积蒸馏水灌胃。喂养8周后取材,采用Masson染色法测定心肌组织胶原蛋白分布及含量。Western blot法检测心肌组织中1型胶原蛋白(Collagen 1)、Ⅲ型胶原蛋白(Collagen 3)、p38MAPK、NF-κB p65蛋白的表达水平。结果:益心附葶饮可降低心衰大鼠心肌纤维组织表达面积。与假手术组比较,模型组大鼠心脏组织Collagen 1、Collagen 3、p38MAPK、NF-κB p65蛋白表达升高,差异有统计学意义(均P<0.05)。中药组Collagen 1、Collagen 3、NF-κB p65蛋白表达低于模型组,差异有统计学意义(均P<0.05)。中药组与模型组p38MAPK蛋白表达量比较,差异无统计学意义(P>0.05)。结论:益心附葶饮通过下调Collagen 1、Collagen 3蛋白表达和抑制NF-κB p65信号通路介导的炎症反应,抑制压力性心力衰竭大鼠心肌纤维化。
Abstract:
Objective:To study the effect and mechanism of Yixin Futing Yin on myocardial fibrosis in rats with congestive heart failure caused by abdominal aortic constriction.Methods:The abdominal aorta constriction method was used to construct rat model of heart failure.The 30 rats were successfully modeled and randomly divided into model group and traditional Chinese medicine group,and 15 rats with threading and no ligation were divided into sham operation group.The traditional Chinese medicine group was given Yixin Futing Yin 3.5 g/(kg·d).The sham operation group and the model group were given equal volume of distilled water.After 8 weeks of feeding,the material was taken,and Masson staining method was used to determine the distribution and content of myocardial tissue collagen.Western blot method was used to detect Collagen 1,Collagen 3,the expression level of p38MAPK,NF-κB p65 proteins.Results:Yixin Futing Yin could reduce the expression area of myocardial fibrous tissue in rats with heart failure.Compared with the sham operation group,the expression of Collagen 1,Collagen 3,p38MAPK and NF-κB p65 proteins in the heart of the model group increased,difference statistically significant(all P<0.05).The expression of Collagen 1,Collagen 3,and NF-κB p65 protein in the traditional Chinese medicine group were lower than those in the model group,difference statistically significant(all P<0.05).There was no significant difference in the expression of p38MAPK protein between the traditional Chinese medicine group and the model group(P>0.05).Conclusion:Yixin Futing Yin inhibited myocardial fibrosis in rats with stress heart failure by down-regulating Collagen 1 and Collagen 3 protein expression and inhibiting inflammatory response mediated by NF-κB p65 signaling pathway.
