摘要:
目的:探讨紫草素在结肠癌细胞5-氟尿嘧啶(5-FU)耐药中的作用及Src-FAK通路的介导作用。方法:以结肠癌HT-29细胞为研究对象构建5-FU耐药细胞HT-29/DR、紫草素处理细胞。克隆形成实验检测细胞克隆形成能力; MTT法检测细胞活力; 蛋白印迹实验检测p-Src、p-FAK、E-cadherin及Fibronectin蛋白表达。结果:HT-29/5-FU细胞构建成功,耐药指数为6.5。与HT-29细胞比较,5-FU处理后HT-29/5-FU细胞克隆形成率显著升高,差异有统计学意义(P<0.01)。5 μmol/L紫草素与10 μmol/L紫草素均可降低HT-29/5-FU和HT-29细胞的存活率。5 μmol/L紫草素与10 μmol/L紫草素均能显著降低5-FU对HT-29/5-FU细胞的IC50,干预前后比较,差异有统计学意义(P<0.01)。5 μmol/L紫草素与10 μmol/L紫草素均能降低HT-29/5-FU细胞p-Src、p-FAK及Fibronectin蛋白表达水平,且能上调E-cadherin蛋白表达,干预前后比较,差异有统计学意义(P<0.01)。结论:紫草素通过抑制Src-FAK通路抑制上皮间质转化,从而缓解结肠癌细胞5-FU耐药。
