摘要:
目的:探讨独活寄生汤通过调控IL-17/NF-κB信号通路治疗膝骨关节炎(KOA)的作用机制。方法:将大鼠随机分为正常组、模型组、独活寄生汤组和双醋瑞因组,采用关节腔注射碘乙酸钠溶液建立KOA模型。确认造模成功后,分别采用药物干预30 d,观察大鼠膝关节软骨病理形态变化。酶联免疫吸附试验(ELISA)检测大鼠血清中白细胞介素17(IL-17)和肿瘤坏死因子α(TNF-α)的表达。Real-time PCR和免疫组化法检测软骨核因子κB p65(NF-κB p65)、肿瘤坏死因子受体相关因子6(TRAF6)、NF-κB激活剂1(ACT1)mRNA和蛋白表达水平。结果:独活寄生汤组大鼠膝关节软骨表面稍不平整,软骨细胞数量减少,排列较规则,软骨基质轻染。与正常组比较,模型组大鼠血清IL-17、TNF-α表达水平升高(均P<0.01)。与模型组比较,独活寄生汤组大鼠血清IL-17、TNF-α表达下降(均P<0.05)。与正常组比较,模型组TRAF6 mRNA表达升高,NF-κB p65、ACT1 mRNA表达升高(均P<0.01)。与模型组比较,独活寄生汤组NF-κB p65、ACT1、TRAF6 mRNA表达降低(均P<0.05)。与正常组比较,模型组大鼠软骨NF-κB p65、TRAF6和ACT1蛋白表达水平升高(均P<0.01)。与模型组比较,独活寄生汤组大鼠软骨NF-κB p65、TRAF6、ACT1的蛋白表达降低(均P<0.01)。结论:独活寄生汤通过调节IL-17/NF-κB信号通路,降低IL-17、TNF-α促炎因子表达,有效缓解KOA模型大鼠症状。
Abstract:
Objective:To explore mechanism of Duhuo Jisheng decoction in the treatment of knee osteoarthritis by regulating IL-17/NF-κB signal pathway.Methods:Rats were randomly divided into normal group,model group,Duhuo Jisheng decoction group and diacerein group.The KOA model was established by intra-articular injection of MIA solution.After continuous intervention for 30 days with different drugs,observing the pathological changes of articular cartilage of knee joint in rats.Real-time fluorescence quantitative polymerase chain reaction was combined with immunohistochemistry to detect the mRNA and protein levels of NF-κB p65,TRAF6 and ACT1 in cartilage.Results:In Duhuo Jisheng decoction group the surface of knee cartilage was slightly uneven the number of chondrocytes decreased,the arrangement regular,and the cartilage matrix lightly stained.Compared with the normal group,the expression levels of IL-17 and TNF-α in serum of the model group were increased(all P<0.01).Compared with the model group,the expression of IL-17 and TNF-α in serum of rats in Duhuo Jisheng decoction group decreased(all P<0.05).Compared with the normal group,the mRNA expression of TRAF6,NF-κB p65 and ACT1 in the model group increased significantly(all P<0.01).Compared with the model group,the mRNA expression of NF-κB p65,ACT1 and TRAF6 in Duhuo Jisheng decoction group decreased significantly(all P<0.05).Compared with the normal group,the protein expression levels of NF-κB p65,TRAF6 and ACT1 in the cartilage of the model group were significantly higher(all P<0.01).Compared with the model group,the protein expression levels of NF-κB p65,TRAF6 and ACT1 in the cartilage of rats in Duhuo Jisheng decoction group decreased significantly(all P<0.01).Conclusion:Duhuo Jisheng decoction can reduce the expression of IL-17 and TNF-α proinflammatory factors by regulating IL-17/NF-κB signal pathway,and effectively alleviate symptom of KOA model rats.
