摘要:
目的:探究温化方调控转录因子κB(NF-κB)信号通路对大鼠膝骨关节炎(KOA)的影响。方法:将SD大鼠随机分为对照组、模型组、温化方组、双氯芬酸钠组、抑制剂组,各10只。除对照组外,其余各组大鼠采用弗氏不完全佐剂乳化的牛Ⅱ型胶原蛋白诱导KOA大鼠模型。造模成功后开始给药,模型组给予0.9%氯化钠灌胃,观察组给予温化方灌胃,双氯芬酸钠组给予双氯芬酸钠灌胃,抑制剂组给予NF-κB抑制剂(吡咯烷二硫代氨基甲酸盐)灌胃,对照组大鼠自由饮水和进食。比较各组HE染色情况,大鼠体重、关节炎指数、关节肿胀度变化情况,酶联免疫吸附试验检测血清IL-1β、IL-6、TNF-α水平,荧光定量PCR法和Western blot检测大鼠关节滑膜组织中NF-κB信号通路相关基因和蛋白表达水平。结果:与模型组比较,对照组、温化方组、双氯芬酸钠组及抑制剂组的大鼠软骨损伤、纤维增生较低。与模型组比较,对照组、温化方组、双氯芬酸钠组、抑制剂组的大鼠体重较高,关节炎指数、关节肿胀度较低(均P<0.05)。与模型组比较,对照组、温化方组、双氯芬酸钠组、抑制剂组IL-1β、IL-6 和TNF-α水平较低(P<0.05)。与模型组比较,对照组、温化方组、双氯芬酸钠组、抑制剂组的NF-κB基因、蛋白表达水平均较低(P<0.05)。结论:温化方通过抑制NF-κB信号通路缓解大鼠KOA病情。
Abstract:
Objective:To explore effect of Wenhua prescription regulating transcription factor κB(NF-κB)signaling pathway on knee osteoarthritis(KOA)in rats.Methods:SD rats were randomly divided into blank control group,model group,Wenhua prescription group,diclofenac sodium group and inhibitor group,10 rats in each group.Except for the blank control group,the other groups of rats were induced by bovine type Ⅱ collagen emulsified with Freund's incomplete adjuvant to induce KOA rat model.After successful modeling,the drug was administered.The model group was given normal saline by gavage,the observation group given Wenhua prescription,the diclofenac sodium group given diclofenac sodium,and the inhibitor group given NF-κB inhibitor(pyrrolidine dithiocarbamate)by gavage,and the rats in the blank control group drank water and ate freely.The HE staining,changes of body weight,arthritis index and joint swelling in each group were compared.The serum IL-1β,IL-6 and TNF-α were detected by enzyme-linked immunosorbent assay.Fluorescence quantitative PCR and Western blot were used to detect the expression levels of NF-κB signaling pathway-related genes and proteins in the synovial tissue of rats.Results:Compared with the model group,the cartilage damage and fibrosis in the control group,Wenhua prescription group,diclofenac sodium group and inhibitor group were lower.The rats had higher body weight,lower arthritis index and lower joint swelling(all P<0.05).Compared with the model group,the control group,Wenhua prescription group,diclofenac sodium group and inhibitor group had lower lever of IL-1β,IL-6 and TNF-α(all P<0.05).Compared with the model group,the control group,Wenhua prescription group,diclofenac sodium group and inhibitor group had lower expression levels of NF-κB gene and protein(all P<0.05).Conclusion:Wenhua prescription alleviates KOA in rats by inhibiting NF-κB signaling pathway.
