摘要:
目的:探讨化痰祛瘀方对心房颤动大鼠的治疗作用及对血清中血管细胞黏附分子-1(VCAM-1)、血管性血友病因子(vWF)的影响。方法:将60只雄性SD大鼠随机分组,喂饲高脂饮食并注射乙酰胆碱氯化钙混合液制备房颤痰浊血瘀证大鼠模型。造模7 d后,连续灌胃化痰祛瘀方2周,观察各组大鼠房颤持续时间、透射电镜下心房肌细胞超微结构以及血清中VCAM-1、vWF水平。结果:中药高浓度组、中药中浓度组、中药低浓度组、维拉帕米组大鼠房颤持续时间均短于模型组,差异有统计学意义(均P<0.05)。透射电镜下,模型组大鼠心房肌细胞的肌小节断裂,心肌细胞肌丝溶解断裂,线粒体肿胀、增多,嵴断裂溶解,呈空泡样改变。中药中浓度组大鼠心房肌细胞的肌小节相对完整,肌丝稍模糊,线粒体肿胀,偶见线粒体溶解,结构相对完整。中药高浓度组、中药中浓度组大鼠血清VCAM-1浓度均低于模型组,差异有统计学意义(均P<0.05)。中药高浓度组、中药中浓度组、中药低浓度组、维拉帕米组大鼠血清vWF均低于模型组,差异有统计学意义(均P<0.05)。结论:化痰祛瘀方可使房颤痰浊血瘀证大鼠房颤持续时间缩短,延缓心房肌细胞纤维化,降低血清中VCAM-1、vWF浓度,对血管内皮细胞有保护作用。
Abstract:
Objective:To investigate therapeutic effects of Huatan Quyu formula on atrial fibrillation rats and effects on serum levels of vascular cell adhesion molecule-1(VCAM-1)and vascular hemophilia factor(vWF).Methods:60 male SD rats were randomly divided.The rats were fed with high-fat diet and injected with acetylcholine calcium chloride mixture to prepare model of atrial fibrillation with phlegm and blood stasis.After 7 days of modeling,Huatan Quyu formula was given intragastric administration for 2 weeks.Changes in duration of atrial fibrillation,ultrastructure of atrial myocytes serum VCAM-1 and vWF were observed under transmission electron microscope.Results:The duration of atrial fibrillation were shorter in the high concentration group,the medium concentration group,the low concentration group and the verapamil group than that in the model group,difference statistically significant(all P<0.05).In the model group atrial myocytes myocytes were broken,myofilaments of cardiac myocytes lysed and broken,mitochondria swollen and increased,cristae broken and dissolved,and there were vacuolar-like changes.In the medium concentration group,the myocytes were relatively intact,myofilaments slightly blurred,mitochondria swollen,mitochondria occasionally dissolved,and the structure was relatively intact.Compared with the model group,the serum VCAM-1 concentration of rats in the high concentration group and the medium concentration group decreased,difference statistically significant(all P<0.05).Compared with the model group,the serum vWF concentration of rats in the high concentration group,the medium concentration group,the low concentration group and the verapamil group were decreased,difference statistically significant(all P<0.05).Conclusion:Huatan Quyu formula can shorten the duration of atrial fibrillation,delay atrial myocyte fibrosis,reduce serum VCAM-1 and vWF concentration and protect vascular endothelial cells in rats with atrial fibrillation of Tanzhuo Xueyu syndrome.
