摘要:
目的:运用网络药理学方法探讨桃红四物汤治疗糖尿病周围神经病变的作用机制。方法:根据中药系统药理学数据库及分析平台(TCMSP)筛选桃红四物汤药物活性成分和靶点。根据Gene Cards、OMIM数据库筛选糖尿病周围神经病变(DPN)靶点基因,构建韦恩图,得到桃红四物汤治疗DPN的作用靶点,应用Cytoscape 3.7.2软件构建药物-成分-靶点-疾病网络。应用R软件进行基因本体(GO)功能注释和东京基因组百科全书(KEGG)通路富集分析。结果:共筛选出桃红四物汤43种有效成分,212个作用靶点,4662个DPN相关靶点,桃红四物汤干预DPN主要涉及150个靶点基因。富集出生物过程155条、信号通路166条。结论:桃红四物汤可能通过调节AKT1、JUN、MAPK1等靶点,调控AGE-RAGE信号通路、流体剪切应力与动脉粥样硬化、IL-17信号通路等多种途径治疗DPN。
Abstract:
Objective:To investigate the mechanism of Taohong Siwu decoction treatment in diabetic peripheral neuropathy(DPN)based on network pharmacology.Methods:The active ingredients and targets of Taohong Siwu decoction were screened through traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP).The Gene Cards database and OMIM database were applied to collect target genes related to DPN.The Venn diagram was constructed to collect the targets of Taohong Siwu decoction in the treatment of DPN.The drug-components-targets-disease network was constructed by Cytoscape 3.7.2 software.The gene oesthetics(GO)function annotation and kyoto encyclopedia of genes and genomes(KEGG)signaling pathway enrichment analysis were performed using R software.Results:The 43 active components and 212 effective targets of Taohong Siwu decoction were searched.4662 effective targets of DPN were predicted.Among these genes,150 genes were concluded in Taohong Siwu decoction treating DPN process and these targets mainly enriched in 155 biological processes and 166 signaling pathways.Conclusion:Taohong Siwu decoction may regulate AGE-RAGE signaling pathway in diabetic complications,fluid shear stress and atherosclerosis,IL-17 signaling pathway signaling pathways to treat DPN through regulating AKT1,JUN,MAPK1.
