摘要:
目的:观察参七虫草方对特发性肺纤维化(IPF)大鼠肺组织缺氧诱导因子(HIF-1α)、血管内皮生长因子(VEGF)、促进磷酸肌3激酶(PI3K)、AKT mRNA的影响。方法:SPF级SD大鼠128只随机分为四组,空白组、模型组、氯沙坦组、参七虫草组,各32只。采用一次性气管滴注博来霉素的方法建立IPF大鼠模型,常规法检测四组大鼠的第7、14、21、28天体重; HE、Masson染色观察四组大鼠的第7、14、21、28天肺组织病理变化; RT-PCR检测各组大鼠肺组织第7、14、21、28天HIF-1α、VEGF、PI3K、AKT mRNA的表达水平。结果:模型组、氯沙坦组及参七虫草组大鼠在镜下均有不同程度的炎性浸润、胶原纤维沉积; 模型组、氯沙坦组及参七虫草组第7、14、21、28天肺组织VEGF、HIF-1α高表达,PI3K、AKT mRNA、 HIF-1α、VEGF、PI3K、AKT mRNA表达水平均显著高于空白组(P<0.05),且随时间推进,差异越显著; 在相同时间点,氯沙坦组及参七虫草组HIF-1α、VEGF、PI3K、AKT mRNA表达水平显著低于模型组(P<0.05),氯沙坦组与参七虫草组比较差异无统计学意义(P>0.05)。结论:参七虫草方可通过抑制大鼠HIF-1α、VEGF、PI3K、AKT mRNA等促血管生成因子的生成而发挥抗纤维化作用。
Abstract:
Objective:To observe the dynamic effects of Shenqi Chongcao formula on HIF-1α,VEGF,PI3K and AKT mRNA in the lung tissue of IPF rats.Methods:The 128 SPF-grade SD rats were randomly and equally divided into 4 groups of 32 rats each:blank group,model group,cloxacin group and Shenqi Chongcao group.A rat model of IPF was established using a single tracheal drip of bleomycin.The routine method of measuring body mass on days 7,14,21 and 28 in four groups of rats.And HE and Masson staining were used to observe the histopathological changes in the lungs of the four groups of rats.The expression levels of PI3K,AKT,HIF-1α and VEGF mRNA were measured by RT-PCR in the lung tissue of each group of rats.Results:Rats in the model group,the cloxacin group and the Shenqi Chongcao group all showed varying degrees of inflammatory infiltration and collagen fibril deposition microscopically.The expression levels of VEGF,PI3K,AKT and HIF-1α mRNA in the lung tissues of the model group,the cloxacin group and the Shenqi Chongcao group at days 7,14,21 and 28 were significantly higher than those in the blank group(P<0.05),and the differences became more significant as time progressed.At the same time points,the expression levels of PI3K,AKT,HIF-1α and VEGF mRNA were significantly lower in the cloxacin group as well as the Shenqi Chongcao group(P<0.05),and the differences between the cloxacin group and Shenqi Chongcao group were not statistically significant(P>0.05).Conclusion:Shenqi Chongcao formula can exert anti-fibrotic effects by inhibiting the production of pro-angiogenic factors such as HIF-1α,VEGF,PI3K and AKT mRNA in rats.
