摘要:
目的:探讨基于Janus激酶2(JAK2)/信号转导和转录激活剂(STAT3)信号通路探讨香仁胃舒汤对慢性非萎缩性胃炎(CNAG)大鼠的作用机制。方法:以化学刺激联合饥饱失常法构建CNAG大鼠模型,将造模成功后的40只存活大鼠随机分为五组,分别为模型组、奥美拉唑组和香仁胃舒汤高、中、低剂量组,每组8只。另选取8只正常SD大鼠作为正常组。奥美拉唑组给予60 mg/kg奥美拉唑溶液3 ml灌胃,香仁胃舒汤高、中、低剂量组分别给予270、180、90 mg/kg香仁胃舒汤3 ml灌胃,正常组和模型组提供等量0.9%氯化钠溶液连续灌胃4周。治疗4周后用苏木素-伊红染色法分析大鼠胃黏膜组织形态变化及病理评分; 用酶联免疫吸附法测定血清中P-JAK2、JAK2、STAT3、白细胞介素-8(IL-8)、白细胞介素-1β(IL-1β)水平; 用蛋白免疫印迹法测定大鼠胃组织中P-JAK2、JAK2、STAT3、IL-8、IL-1β蛋白表达。结果:高剂量香仁胃舒汤能降低大鼠胃体及胃窦黏膜组织病理评分、血清P-JAK2、JAK2、STAT3、IL-8、IL-1β水平及胃黏膜组织P-JAK2、JAK2、STAT3、IL-8、IL-1β蛋白表达(P<0.05)。结论:香仁胃舒汤可在一定程度上修复CNAG大鼠胃黏膜损伤,其机制与抑制JAK2/STAT3信号通路,降低P-JAK2、JAK2、STAT3、IL-8及IL-1β表达有关。
Abstract:
Objective:To explore the mechanism of Xiangren Weishu decoction on CNAG rats based on Janus kinase 2(JAK2)/signal transducer and activator of transcription(STAT3)signaling pathway.Methods:The CNAG rat model was established by chemical stimulation combined with hunger and satiety disorder,and 40 surviving rats after successful modeling were randomly divided into 5 groups,namely model group,omeprazole group and Xiangren Weishu decoction high,medium,and low-dose group,there were 8 rats in each group.Another 8 normal SD rats were selected as the normal group.The omeprazole group was given 60 mg/kg omeprazole solution 3 ml by gavage,and the high,medium and low-dose groups of Xiangren Weishu decoction were given 270,180,90 mg/kg Xiangren Weishu decoction 3 ml by gavage,the normal group and the model group were given the same volume of normal saline by continuous gavage for 4 weeks.After 4 weeks of treatment,HE staining was used to analyze the morphological changes and pathological scores of gastric mucosa in rats; ELISA was used to determine serum P-JAK2,JAK2,STAT3,IL-8,IL-1β levels; P-JAK2,JAK2,STAT3,IL-8,IL-1β proteins express in rat gastric tissue were determined by Western blot.Results:The high-dose group of Xiangren Weishu decoction can reduce the histopathological score of gastric body and gastric antrum mucosa,serum P-JAK2,JAK2,STAT3,IL-8,IL-1β levels and gastric mucosa tissue P-JAK2,JAK2,STAT3,IL-8,IL-1β protein expression(P<0.05).Conclusion:Xiangren Weishu decoction can repair gastric mucosal injury in CNAG rats to a certain extent,and its mechanism is related to inhibiting the opening of JAK2/STAT3 signaling pathway and reducing the expressions of P-JAK2,JAK2,STAT3,IL-8 and IL-1β.
