摘要:
目的:研究黄芩素在伊立替康抑制结肠癌细胞增殖中的作用及相关机制。方法:用结肠癌HT-29细胞作为研究对象。以50、100、200 μmol/L黄芩素、5 μmol/L伊立替康分别处理结肠癌HT-29细胞。MTT实验和平板克隆实验检测细胞增殖和克隆形成能力; Western blotting法实验检测Src激酶、Yes相关蛋白(YAP)及大肿瘤抑制因子激酶1(LATS1)磷酸化蛋白表达; 免疫荧光实验检测细胞YAP表达分布。结果:50 μmol/L黄芩素组、100 μmol/L黄芩素组、200 μmol/L黄芩素组均能增加伊立替康对HT-29细胞的增殖和克隆抑制作用。50 μmol/L黄芩素组、100 μmol/L黄芩素组、200 μmol/L黄芩素组均可下调结肠癌HT-29细胞p-Src蛋白表达,上调p-YAP、p-LATS1蛋白表达。200 μmol/L黄芩素组能增加结肠癌HT-29细胞的细胞质YAP分布。结论:黄芩素通过抑制Src-YAP信号通路增强伊立替康对结肠癌细胞的抑制作用。
Abstract:
Objective:To study effect of baicalein on irinotecan's inhibition of colon cancer cell proliferation and related mechanisms.Methods:Colon cancer HT-29 cells were used as research objects.Cells were treated with 50 μmol/L baicalein,100 μmol/L baicalein,200 μmol/L baicalein,5 μmol/L irinotecan.MTT assay and plate cloning assay were used to detect cell proliferation and clone formation.Western blotting assay was used to detect expression of Src kinase,Yes-related protein(YAP)and large tumor suppressor kinase 1(LATS1)phosphorylated protein.Immunofluorescence assay was to detect distribution of YAP expression in cells.Results:50 μmol/L baicalein,100 μmol/L baicalein,200 μmol/L baicalein could increase inhibitory effect of irinotecan on proliferation and colony formation of HT-29 cells.50 μmol/L baicalein,100 μmol/L baicalein,200 μmol/L baicalein could down-regulate p-Src protein expression and up-regulate p-YAP,p-LATS1 protein expression in HT-29 cells.200 μmol/L baicalein can increase distribution of cytoplasmic YAP in HT-29 cells.Conclusion:Baicalein may enhance the inhibitory effect of irinotecan on colon cancer cell proliferation by inhibiting Src-YAP signaling pathway.
