摘要:
目的:观察支气管哮喘大鼠模型肺组织中嗜酸性粒细胞(EOS)的凋亡及其调控蛋白Fas、Bcl-2的表达,探析加味过敏煎对模型的干预作用。方法:将50只大鼠随机分为空白组、模型组、加味过敏煎组、激素组、加味过敏煎联合激素组,每组10只。用氢氧化铝/卵蛋白[Al(OH)3/OVA]致敏,雾化吸入含OVA的0.9%氯化钠溶液激发,建立哮喘模型。各治疗组均给予相应药物连续灌胃14 d,并在灌胃治疗后雾化含OVA的0.9%氯化钠溶液激发,每次雾化20 min,连续雾化7 d。模型组与空白组均用0.9%氯化钠溶液进行灌胃与雾化。采用TUNEL免疫荧光染色法观察肺组织中EOS凋亡情况; 免疫印迹法、免疫组化法检测肺组织中Fas、Bcl-2蛋白的表达情况。结果:模型组大鼠肺组织中EOS计数显著增加,EOS凋亡率明显降低,Fas蛋白表达显著降低,Bcl-2蛋白表达显著增高(P<0.05); 治疗后,各治疗组大鼠肺组织中EOS凋亡率均有不同程度的增高,Fas蛋白表达均增高,Bcl-2蛋白表达均降低,其中以加味过敏煎联合激素组变化程度最明显(P<0.05)。结论:加味过敏煎可能通过促进哮喘大鼠肺组织中Fas蛋白表达、抑制Bcl-2蛋白表达,促进EOS凋亡,减轻EOS在肺组织中的浸润与聚集,从而控制哮喘大鼠气道炎性反应,达到治疗支气管哮喘的作用。
Abstract:
Objective:To observe the apoptosis of eosinophils(EOS)and the expression of regulatory proteins Fas and Bcl-2 in the lung tissue of the bronchial asthma rat model and the intervention of Jiawei Guomin decoction.Methods:50 rats were randomly divided into blank group,model group,compound Jiawei Guomin decoction group,hormone group,and Jiawei Guomin decoction combined with hormone group,10 rats in each group.The asthma model was established by sensitizing with Aluminum hydroxide/ovalbumin [Al(OH)3/OVA] and inhaling normal saline containing OVA by atomization.Each treatment group was given corresponding drugs by continuous intragastric administration for 14 days,and after intragastric administration,the normal saline containing ova was atomized for 20 minutes each time for 7 days.Both the model group and the blank group were perfused and atomized with normal saline.TUNEL immunofluorescence staining was used to observe EOS apoptosis in lung tissue.The expression of Fas and Bcl-2 protein in lung tissue was detected by Western blot and immunohistochemistry.Results:The EOS count in the lung tissue of the rats in the model group was significantly increased,the EOS apoptosis rate was significantly decreased,the Fas protein expression was significantly decreased,and the Bcl-2 protein expression was significantly increased(P<0.05).After administration,the EOS apoptosis rate in the lung tissue of rats in each treatment group was increased to varying degrees,the expression of Fas protein was increased,and the expression of Bcl-2 protein was decreased,especially in the Jiawei Guomin decoction combined with hormone group(P<0.05).Conclusion:Jiawei Guomin decoction may control the airway inflammatory response of asthmatic rats and achieve the effect of treating bronchial asthma by promoting the expression of Fas protein,inhibiting the expression of Bcl-2 protein,promoting EOS apoptosis,and reducing the infiltration and aggregation of EOS in the lung tissue of asthmatic rats.
