摘要:
目的:探讨补阳还五汤对急性心肌梗死大鼠内皮祖细胞(EPCs)凋亡的影响及其调控机制。方法:本研究取大鼠36只,心脏采血后分离出EPCs,分为空白组、对照组、中药组、阿托伐他汀组、联合组及抑制剂组,分别给予补阳还五汤、阿托伐他汀钙片、补阳还五汤联合阿托伐他汀及磷酸肌醇3-激酶/蛋白激酶B(PI3K/Akt)通道抑制剂进行干预,于24、48、72 h时进行细胞凋亡和PI3K/Akt通路mRNA表达、蛋白表达及蛋白磷酸化检测。结果:48、72 h时中药组、阿托伐他汀组、联合组及抑制剂组的凋亡率低于对照组,而抑制剂组的凋亡率高于中药组、联合组,差异有统计学意义(均P<0.05)。24 h空白组、对照组的PI3K mRNA和Akt mRNA表达比较,差异无统计学意义(均P>0.05)。72 h中药组、阿托伐他汀组、联合组及抑制剂组的PI3K mRNA和Akt mRNA表达高于对照组,差异有统计学意义(均P<0.05)。各时间点对照组PI3K蛋白表达低于空白组,而中药组、阿托伐他汀组、联合组及抑制剂组高于对照组,差异有统计学意义(均P<0.05)。各时间点中药组、阿托伐他汀组、联合组及抑制剂组Akt蛋白磷酸化水平高于对照组,差异有统计学意义(均P<0.05)。48、72 h抑制剂组的Akt蛋白磷酸化水平低于对照组,差异有统计学意义(均P<0.05)。结论:补阳还五汤可减轻H2O2引起的EPCs凋亡,维持氧化应激状态下的PI3K/Akt通路mRNA、蛋白表达及蛋白磷酸化。在蛋白表达及磷酸化部分,抑制剂不能完全抑制中药含药血清作用,提示补阳还五汤干预氧化应激可能存在除PI3K/Akt通路以外的途径。
Abstract:
Objective:To investigate the effect of Buyang Huanwu decoction on apoptosis of endothelial progenitor cells(EPCs)in rats with acute myocardial infarction(AMI)and its regulatory mechanism.Methods:In this study,36 rats heart blood collection to separated EPCs were divided into blank group,control group,traditional Chinese medicine(TCM)group,atorvastatin group,combined group and inhibitor group.Buyang Huanwu decoction,atorvastatin calcium tablets,Buyang Huanwu decoction combined with atorvastatin and phosphoinositide 3-kinase/protein kinase B(PI3K/Akt)channel inhibitors were given for intervention.Apoptosis and PI3K/Akt pathway mRNA expression,protein expression and protein phosphorylation were detected at 24,48 and 72 h.Results:At 48 h and 72 h,the apoptosis rate of TCM group,atorvastatin group,combined group and inhibitor group were lower than those in the control group,while the apoptosis rate of inhibitor group were higher than those in the TCM group and the combined group,differences statistically significant(all P<0.05).There was no significant difference in PI3K mRNA and Akt mRNA expression between the blank and control groups at 24 h(all P>0.05).At 72 h,PI3K mRNA and Akt mRNA expression in the TCM,atorvastatin,combined,and inhibitor groups were higher than those in the control group,differences statistically significant(all P<0.05).PI3K protein expression was lower in the control group than that in the blank group at each time point,while it was higher in the TCM,atorvastatin,combined,and inhibitor groups than that in the control group,differences statistically significant(all P<0.05).Akt protein phosphorylation levels were higher in the TCM,atorvastatin,combined,and inhibitor groups than in the control group at each time point,differences statistically significant(all P<0.05).The Akt protein phosphorylation level in the inhibitor group was significantly lower than that in the control group at 48 and 72 h(P<0.05).Conclusion:Buyang Huanwu decoction can reduce the apoptosis of EPCs induced by H2O2 and maintain the mRNA and protein expression and protein phosphorylation of PI3K/Akt pathway under oxidative stress.In the protein expression and phosphorylation part,the inhibitor could not completely inhibit the effect of medicated serum of traditional Chinese medicine,suggesting that Buyang Huanwu decoction may have pathways other than PI3K/Akt pathway in intervening oxidative stress.
