摘要:
目的:研究温阳振衰颗粒调控miR-155/p38MAPK途径减轻大鼠心肌缺血再灌注损伤(MIRI)的作用机制。方法:成年雄性SD大鼠随机分为阴性对照组、阴性对照+MIRI组、miR-155抑制剂+MIRI组、miR-155模拟物+MIRI组、对照组、MIRI组、中药组、中药+miR-155抑制剂组,建立MIRI模型,给予温阳振衰颗粒灌胃干预,miR-155抑制剂或模拟物局部注射。比较各组磷酸肌酸激酶同工酶(CK-MB)及乳酸脱氢酶(LDH)含量、miR-155及p-p38MAPK表达、炎症因子肿瘤坏死因子-α(TNF-α)及白介素-6(IL-6)含量。结果:与对照组比较,MIRI组血清CK-MB、LDH含量及心肌p-p38MAPK表达、TNF-α、IL-6含量明显增加,心肌miR-155表达明显减少(均P<0.05); 与MIRI组比较,中药组血清CK-MB、LDH含量及心肌p-p38MAPK表达、TNF-α、IL-6含量明显减少,心肌miR-155表达明显增加(均P<0.05); 与中药组比较,中药+miR-155抑制剂组血清CK-MB、LDH含量及心肌p-p38MAPK表达、TNF-α、IL-6含量明显增加,心肌miR-155表达明显减少(均P<0.05); 与阴性对照+MIRI组比较,miR-155抑制剂+MIRI组心肌损伤加重,miR-155模拟物+MIRI组心肌损伤减轻(均P<0.05)。结论:温阳振衰颗粒通过调控miR-155/p38MAPK途径减轻大鼠MIRI,miR-155能够在心肌MIRI过程中靶向p38MAPK,抑制炎症并减轻心肌损伤。
Abstract:
Objective:To study the effect and mechanism of Wenyang Zhenshuai granule on reducing myocardial ischemia-reperfusion injury(MIRI)via regulating miR-155/p38MAPK pathway.Methods:Adult male SD rats were randomly divided into control group,MIRI group,TCM group,TCM+miR-155 inhibitor group,negative control(NC)group,NC+MIRI group,miR-155 inhibitor+MIRI group,miR-155 mimic+MIRI group.The contents of CK-MB,LDH,the expression of miR-155,p-p38MAPK,the contents of TNF-α and IL-6 were compared among the groups.Results:Compared with the control group,the contents of CK-MB and LDH in serum and the expression of p-p38MAPK,the contents of TNF-α and IL-6 in myocardium of MIRI group significantly increased,and the expression of miR-155 in myocardium significantly decreased(all P<0.05).Compared with MIRI group,the contents of CK-MB and LDH in serum and the expression of p-p38MAPK,the contents of TNF-α and IL-6 in myocardium of TCM group significantly decreased,and the expression of miR-155 in myocardium significantly increased(all P<0.05).Compared with TCM group,the contents of CK-MB and LDH in serum and the expression of p-p38MAPK,the contents of TNF-α and IL-6 in myocardium of TCM+miR-155 inhibitor group significantly increased,and the expression of miR-155 in myocardium significantly decreased(all P<0.05).Compared with NC+MIRI group,myocardial injury was aggravated in miR-155 inhibitor+MIRI group,and myocardial injury was alleviated in miR-155 mimic+MIRI group(all P<0.05).Conclusion:Wenyang Zhenshuai granule can reduce MIRI by regulating miR-155/p38MAPK pathway and miR-155 can inhibit inflammation and reduce injury during MIRI by targeting p38MAPK.
