摘要:
目的:探讨针灸预处理对急性胃黏膜损伤大鼠葡萄糖调节蛋白78(GRP78)/半胱氨酸蛋白水解酶-12(Caspase-12)/CCAAT增强子结合蛋白同源蛋白(CHOP)信号通路的影响。方法:将52只大鼠随机分为正常组、模型组、艾灸预处理组、针刺预处理组,每组13只。正常组、模型组仅固定不作处理; 对针刺预处理组大鼠行针刺处理; 对艾灸预处理组大鼠行足三里穴、中脘穴艾灸,20 min/次,1次/d,连续灸7 d。对模型组、针刺预处理组、艾灸预处理组大鼠进行无水乙醇结合阿司匹林混悬液灌胃建立急性胃黏膜损伤模型。观察大鼠一般情况及胃黏膜损伤组织病理学变化; 酶联免疫吸附实验(ELISA)法检测大鼠血清肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、环氧化酶-2(COX-2)浓度; 免疫组织化学法检测大鼠GRP78、Caspase-12、CHOP水平。结果:模型组大鼠可见上皮细胞结构破坏不完整,胃黏膜组织损伤明显,胃黏膜损伤指数(UI)评分显著高于正常组(P<0.05); 与模型组比较,针刺预处理组、艾灸预处理组大鼠UI指数评分显著降低(均P<0.05)。与正常组比较,模型组大鼠血清TNF-α、IL-6、COX-2浓度显著升高(均P<0.05); 与模型组比较,针刺预处理组、艾灸预处理组大鼠血清TNF-α、IL-6、COX-2浓度显著降低(均P<0.05)。与正常组比较,模型组大鼠GRP78、Caspase-12、CHOP呈强阳性表达,可见大量棕黄色颗粒,针刺预处理组、艾灸预处理组大鼠GRP78、Caspase-12、CHOP结果阳性细胞数量减少,呈弱阳性,可见少量棕黄色颗粒。结论:针灸预处理可减轻急性胃黏膜损伤大鼠炎症反应,其作用机制可能是通过抑制内质网应激,基于GRP78/Caspase-12/CHOP信号通路预防大鼠胃黏膜损伤。
Abstract:
Objective:To investigate the effects of acupuncture pretreatment on the histopathology of gastric mucosa and glucose-regulated protein 78(GRP78)/cysteine protein hydrolase-12(Caspase-12)/CCAAT enhancer-binding protein homologous protein(CHOP)signaling pathways in rats with acute gastric mucosal lesion.Methods:52 rats were randomly divided into normal group,model group,moxibustion pretreatment group,acupuncture pretreatment group,each group consisted of 13 rats.Normal group and model group were fixed without treatment.The rats in the acupuncture pretreatment group were treated with acupuncture.The rats in the moxibustion pretreatment group were subjected to moxibustion at the Zusanli point and the Zhongwan point,20 min each time,moxibustion once a day for 7 days.After pretreatment,rats in the model group,moxibustion pretreatment group and acupuncture pretreatment group were gavaged with anhydrous ethanol combined with aspirin suspension to establish a model of acute gastric mucosal lesion.The general condition of rats and histopathological changes of gastric mucosal injury were observed.The concentrations of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and cyclooxygenase-2(COX-2)in serum of rats were determined by enzyme-linked immunosorbent assay(ELISA).The levels of GRP78,Caspase-12 and CHOP were determined by immunohistochemistry.Results:In the model group,incomplete structural destruction of epithelial cells and significant damage to gastric mucosal tissues were seen,and the UI index score was significantly higher than that of the normal group(P<0.05).Compared with model group,UI index scores of rats in acupuncture pretreatment group and moxibustion pretreatment group were significantly decreased(all P<0.05).Compared with the normal group,the levels of serum TNF-α,IL-6 and COX-2 in the model group were significantly higher(all P<0.05).Compared with the model group,the levels of serum TNF-α,IL-6 and COX-2 concentrations in the acupuncture and moxibustion pretreatment group were significantly lower(all P<0.05).Compared with the normal group,GRP78,Caspase-12 and CHOP in the gastric mucosa of rats in the model group showed strong positive expression and a large number of brownish-yellow granules were visible,while the number of GRP78,Caspase-12 and CHOP positive cells in the acupuncture pretreatment group decreased and were weakly positive,and a small number of brownish-yellow granules were visible.Conclusion:Acupuncture pretreatment can reduce inflammatory response in AGML rats,and its mechanism may be through inhibiting ERS and preventing gastric mucosal injury based on GRP78/Caspase-12/CHOP signaling pathway.
