摘要:
目的:研究头针针刺对血管性痴呆(VD)大鼠行为及海马体胆碱能抗炎通路(CAP)的影响。方法:将94只大鼠随机分为正常组12只、假手术组12只和造模组70只,大鼠VD模型的建立使用改良Pulsinelli四血管阻断法,在造模后第4周随机将造模成功的48只VD大鼠分为模型组、美金刚组、头针组及联合组各12只,美金刚组按体重给予美金刚灌胃,头针组采用针刺百会穴和四神聪治疗,联合组采用头针和美金刚联合治疗,正常组、假手术组和模型组将美金刚组用药量换算为等体积0.9%氯化钠溶液灌胃,比较各组大鼠治疗8周时行为学评价结果。检测各组大鼠海马组织胆碱乙酰转移酶(ChAT)和乙酰胆碱酯酶(AChE)活性以及肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-10(IL-10)、转化生长因子-β(TGF-β)和核因子-κB(NF-κB)表达水平。结果:与造模前比较,造模4周时,模型组、美金刚组、头针组和联合组VD大鼠逃避潜伏期均明显升高(P<0.05),穿越原平台次数均显著下降(P<0.05); 相比于造模4周,治疗8周时各组以上指标显著改善(P<0.05),头针组改善效果显著优于美金刚组,联合组改善效果显著优于美金刚组和头针组(P<0.05)。治疗8周时,与模型组比较,头针组和联合组ChAT、AChE活性显著升高,且联合组ChAT活性高于头针组(P<0.05); 美金刚组、头针组和联合组海马组织TNF-α、IL-1β水平显著降低(P<0.05),IL-10、TGF-β水平显著升高(P<0.05),头针组各指标改善效果均显著优于美金刚组(P<0.05),联合组各指标改善效果均显著优于头针组(P<0.05); 美金刚组、头针组和联合组NF-κB蛋白表达水平显著降低(P<0.05),头针组NF-κB蛋白表达水平低于美金刚组(P<0.05),联合组NF-κB蛋白表达水平低于头针组(P<0.05)。结论:头针针刺治疗可增强VD大鼠ChAT和AChE活性,提升乙酰胆碱代谢水平,增强CAP作用,使海马组织炎症反应有所减轻,大鼠空间学习记忆能力得到明显改善,其作用机制可能与抑制大鼠海马组织NF-κB信号通路有关。
Abstract:
Objective:To study the effect of scalp acupuncture on behavior and Cholinergic anti-inflammatory pathway(CAP)in the hippocampus of vascular dementia(VD)rats.Methods:A total of 94 rats were randomly divided into three groups:normal group(n=12),sham-operation group(n=12),and model group(n=70).VD model was established by modified Pulsinelli four-vessel occlusion method.After four weeks of modeling,48 rats with VD were randomly assigned to four different groups,each containing 12 rats.These groups included the model group,memantine group,scalp acupuncture group,and combination group.The scalp acupuncture group was treated with acupuncture at Baihui point and Sishencong,the combined group was treated with scalp acupuncture and memantine,and the normal group,sham operation group and model group were treated with memantine.The behavioral evaluation results of the rats in each group were compared after 8 weeks of treatment.The activities of Choline Acetyltransferase(ChAT),Acetylcholinesterase(AChE)and Tumor necrosis factor(TNF)in hippocampus were detected Tumor necrosis factor-α(TNF-α),Interleukin-1β(IL-1β),Interleukin-10(IL-10),Transforming growth factor-β(TGF-β)and Nuclear factor the expression level of kappa-B(NF-κB).Results:After 4 weeks of modeling,there was a notable increase in the escape latency of VD rats in the model group,memantine group,scalp acupuncture group,and combined group(P<0.05),and the number of crossing the original platform was significantly reduced(P<0.05); Compared with 4 weeks after modeling,the above indexes in each group were significantly improved at 8 weeks of treatment(P<0.05),the scalp acupuncture group was significantly better than the memantine group,and the combined group was significantly better than the memantine group and the scalp acupuncture group(P<0.05).After 8 weeks of treatment,compared with the model group,the activities of ChAT and AChE in the scalp acupuncture group and the combination group were significantly increased,and the activity of ChAT in the combination group was higher than that in the scalp acupuncture group(P<0.05).The levels of TNF-α and IL-1β in hippocampus of memantine group,scalp acupuncture group and combined group were significantly decreased(P<0.05),and the levels of IL-10 and TGF-β were significantly increased(P<0.05),and the improvement effect of scalp acupuncture group was significantly better than that of memantine group(P<0.05),and the improvement effect of the combined group was significantly better than that of the scalp acupuncture group(P<0.05).The expression of NF-κB protein in memantine group,scalp acupuncture group and combined group was significantly decreased(P<0.05).The expression of NF-κB protein in the scalp acupuncture group was lower than that in the memantine group(P<0.05).The expression of NF-κB protein in the combined group was lower than that in the scalp acupuncture group(P<0.05).Conclusion:Scalp acupuncture can enhance the activities of ChAT and AChE,increase the level of acetylcholine metabolism,enhance the effect of CAP,reduce hippocampal inflammation,and improve spatial learning and memory ability in VD rats,which may be related to the inhibition of NF-κB signaling pathway in the hippocampus.
