摘要:
目的:基于TLR4/MyD88/NF-κB信号通路蛋白表达的观察,探讨针灸对胃黏膜“治未病”的作用机制。方法:将大鼠分为四组:正常组、模型组、预灸组和针预组,预灸组悬灸、针预组针刺“中脘”“足三里”,每穴20 min,1次/d,连续治疗7 d。针灸预处理结束后,模型组、预灸组、针预组制备胃黏膜损伤模型。评估胃黏膜损伤指数(UI),ELISA法检测大鼠血清TNF-α和IL-1β水平,免疫组织化学法和Western blot检测胃黏膜组织TLR4、MyD88、NF-κB p65蛋白表达水平。结果:模型组大鼠UI指数评分高于正常组(P<0.05),观察胃黏膜组织出现明显损伤,有出血灶,提示模型制备成功。与模型组比较,预灸组和针预组大鼠血清TNF-α、IL-1β浓度和TLR4、MyD88、NF-κB p65蛋白表达水平显著降低(P<0.05)。结论:针灸预处理对胃黏膜损伤可以起到预防作用,其作用机制可能是通过对TLR4/MyD88/NF-κB p65信号通路蛋白表达水平的调控,对大鼠胃黏膜损伤的炎性反应起抑制作用,从而达到对胃黏膜组织的保护作用。
Abstract:
Objective:To investigate the mechanism of acupuncture on the treatment of gastric mucosa based on the expression of TLR4/MyD88/NF-κB signaling pathway.Methods:The rats were divided into four groups:normal group,model group,pre-moxibustion group and acupuncture group,and acupuncture group with suspension moxibustion and acupuncture in the pre-moxibustion group and acupuncture Zusanli for 20 min per acupoint,1 time/d,and continuous treatment for 7 days.After the acupuncture pretreatment,the model group,the pre-moxibustion group and the needle pre-group prepared a model of gastric mucosal injury.Gastric mucosal damage index(UI)was assessed,serum TNF-α and IL-1β levels were measured by ELISA and TLR4,MyD88,NF-κB p65 protein expression levels were measured by immunohistochemistry and Western blot.Results:The UI index score of rats in the model group was higher than that in the normal group(P<0.05),and obvious damage and bleeding foci were observed in the gastric mucosal tissue,indicating that the model was successfully prepared.Compared with the model group,serum TNF-α,IL-1β concentrations,TLR4,MyD88 and NF-κB p65 protein expression levels in the pre-moxibustion group and the needle pre-group were significantly reduced(P<0.05).Conclusion:Acupuncture pretreatment can play a preventive role in gastric mucosal injury,and its mechanism of action may be:through the regulation of TLR4/MyD88/NF-κB p65 signaling pathway protein expression level,it inhibits the inflammatory response of gastric mucosal injury in rats,thereby achieving a protective effect on gastric mucosal tissue.
