摘要:
目的:探讨通窍活血汤对血管性痴呆大鼠海马组织miR-9、miR-124的影响。方法:取健康SD大鼠为实验对象,采用缺血再灌注配合永久性结扎双侧CCA法制备模型,将其分为假手术组、模型组、中药组和安慰剂组,每组12只。中药组大鼠予通窍活血汤灌胃,安慰剂组大鼠予同体积0.9%氯化钠溶液灌胃,分别持续灌胃4周,1次/d; 假手术组及模型组大鼠不予干预。于造模后第1天及干预28 d测定大鼠卒中指数评分、神经病学症状评分及行为学检测,用RT-qPCR法检测大鼠海马组织miR-9、miR-124含量,苏木素-伊红染色法观察大鼠海马病理学结构情况。结果:通过HE染色可知,假手术组大鼠海马组织神经元结构完整,细胞核染色均匀; 模型组及安慰剂组大鼠海马组织神经元结构残缺,神经元丢失,细胞核染色不均匀,形态固缩; 中药组大鼠海马组织神经元结构相对完整,神经元有少量丢失,细胞核染色程度减低,细胞核固缩形态相对减少; 通窍活血汤治疗血管性痴呆大鼠可降低其卒中指数及神经病学症状评分(P<0.05),缩短其逃避潜伏期耗时,增加穿越平台次数(P<0.05),其海马组织miR-9、miR-124表达水平下降(P<0.05)。结论:通窍活血汤可以改善血管性痴呆大鼠空间学习及记忆障碍,其机制可能是通过抑制海马区miR-9、miR-124的过度表达有关。
Abstract:
Objective:To investigate the effects of Tongqiao Huoxue decoction on miR-9 and miR-124 in hippocampus of vascular dementia rats.Methods:In this experiment,healthy SD rats were selected as experimental subjects.They were divided into sham operation group,model group,traditional Chinese medicine group and placebo group,with 12 rats in each group.The rats in the Chinese medicine group were given Tongqiao Huoxue decoction and the rats in the placebo group were given 0.9% sodium chloride solution for continuous gavage for 4 weeks,once a day.The stroke index score,neurological symptom score and behavioral detection were determined on the 1st day after modeling and the 28th day after intervention.The contents of miR-9 and miR-124 in the hippocampus were detected by RT-qPCR.The pathological structure of the hippocampus was observed by hematoxylin-eosin staining.Results:HE staining showed that the hippocampal neurons in the sham operation group were intact and the nucleus staining was uniform.In the model group and the placebo group,the hippocampal neuron structure was incomplete,the neuron was lost,the nucleus staining was uneven,and the shape was pinched.In the Chinese medicine group,the hippocampal neuron structure was relatively complete with a small amount of neuron loss,the degree of nuclear staining was reduced,and the form of nuclear shrinkage was relatively reduced.The treatment of Tongqiao Huoxue decoction in vascular dementia rats could reduce their stroke index and neurological symptom score(P<0.05),shorten their escape latency time,and increase the number of crossing platforms(P<0.05).The expression levels of miR-9 and miR-124 in hippocampus were decreased(P<0.05).Conclusion:Tongqiao Huoxue decoction can improve spatial learning and memory impairment in vascular dementia rats,and the mechanism may be related to inhibiting the overexpression of miR-9 and miR-124 in hippocampus.
