摘要:
目的:观察嗅三针对血管性痴呆(VD)大鼠行为学、海马组织病理形态、Keap1/Nrf2/ARE信号通路关键蛋白的影响。方法:60只SD大鼠随机分为假手术组、模型组、嗅三针组、抑制剂组,每组15只。采用双侧颈总动脉结扎术建立VD模型。嗅三针给予带电干预。采用Morris水迷宫检测大鼠行为学变化,HE染色检测海马CA1区组织形态变化,免疫荧光法检测海马CA1区Nrf2蛋白表达情况,Western blotting检测HO-1、NQO1蛋白表达情况。结果:模型组、抑制剂组大鼠寻找目标平台游泳的距离明显大于假手术组、嗅三针组。空间探索试验结果中,假手术组、嗅三针组穿越平台区次数、平台区游泳距离明显多于模型组、抑制剂组。模型组病理形态中细胞排列紊乱,坏死数量多于假手术组。嗅三针组海马区病理形态的细胞形态清晰、排列整齐,优于模型组。抑制剂组海马区细胞排列不齐,数目少于嗅三针组; 嗅三针组Nrf2蛋白表达高于模型组、抑制剂组。嗅三针组大鼠海马组织HO-1、NQO1表达高于模型组、抑制剂组,差异有统计学意义(均P<0.05)。结论:嗅三针能够改善VD大鼠认知障碍,减少海马损伤,通过激活Nrf2,调节HO-1、NQO1蛋白表达,发挥对抗氧化应激作用。
Abstract:
Objective:To observe effect of olfactory three needles on behavior,pathological morphology of hippocampus and key protein of Keap1/Nrf2/ARE signaling pathway in mice with vascular dementia(VD).Methods:Sixty SD mice were randomly divided into sham operation group,model group,olfactory three needles group and inhibitor group,with 15 mice in each group.VD model was established by ligation of bilateral common carotid arteries.Olfactory three needles were given electric intervention.Morris water maze was applied to detect behavioral changes of mice.HE staining was applied to detect morphological changes of hippocampal CA1 area.Immunofluorescence method was applied to detect expression of Nrf2 protein in hippocampal CA1 region.The expression of HO-1 and NQO1 protein were detected by Western blotting.Results:The swimming distance of mice in the model group and inhibitor group to find target platform were significantly greater than that in the sham operation group and olfactory three needles group.In the results of space exploration test,the number of times of crossing platform area and swimming distance of the platform area in the sham operation group and olfactory three needles group were significantly more than those in the model group and the inhibitor group.The cell arrangement in the model group was disordered and the number of necrosis higher than that in the sham operation group.The pathological morphology of the cells in the hippocampus of the olfactory three needles group was clear and orderly better than that in the model group.The cells in the hippocampus of the inhibitor group were disordered,and the number was reduced in the olfactory three needles group group.The expression of Nrf2 protein in the olfactory three needles group group was higher than that in model group and inhibitor group.The expressions of HO-1 and NQO1 in hippocampal tissue of rats in the olfactory three needles group were higher than those in the model group and inhibitor group,differences statistically significant(all P<0.05).Conclusion:The olfactory three needles can improve cognitive impairment and reduce hippocampal damage in VD mice,which may be due to its anti-oxidative stress effect by activating Nrf2 and regulating expression of HO-1 and NQO1 proteins.
