摘要:
目的:探讨中药土贝母主要成分土贝母皂苷甲(TBMS Ⅰ)通过激活Egr1/p21信号通路对乳腺癌细胞MCF-7凋亡的影响。方法:体外培养MCF-7细胞,并把细胞分为空白组、TBMS Ⅰ高剂量组(20 μg/ml)、TBMS Ⅰ中剂量组(15 μg/ml)、TBMS Ⅰ低剂量组(10 μg/ml),使用对应浓度的TBMS Ⅰ干预MCF-7细胞。干预24、48、72 h后,使用CCK-8法检测细胞增殖情况。干预MCF-7细胞48 h后,采用流式细胞仪检测各组细胞凋亡、细胞周期分布情况。采用Western blotting、qRT-PCR检测各组细胞Egr1、p21蛋白及mRNA表达。结果:干预24、48、72 h后,TBMS Ⅰ高剂量组、TBMS Ⅰ中剂量组的MCF-7细胞得到抑制,且抑制率呈时间与剂量依赖性。干预48 h后,TBMS Ⅰ高剂量组、TBMS Ⅰ中剂量组、TBMS Ⅰ低剂量组MCF-7细胞的凋亡率高于空白组,差异有统计学意义(均P<0.05)。TBMS Ⅰ高剂量组、TBMS Ⅰ中剂量组、TBMS Ⅰ低剂量组G0/G1期细胞占比低于空白组,G2/M期细胞高于空白组,差异有统计学意义(均P<0.05)。TBMS Ⅰ高剂量组、TBMS Ⅰ中剂量组、TBMS Ⅰ低剂量组Egr1及p21蛋白、mRNA表达均高于空白组,差异有统计学意义(均P<0.05)。结论:TBMS Ⅰ能够激活乳腺癌细胞MCF-7中Egr1/p21信号通路,诱导Egr1、p21表达,引起MCF-7细胞G2/M期周期阻滞,诱导MCF-7细胞凋亡。
Abstract:
Objective:To investigate effect of main ingredient of Chinese medicine paniculate bolbostemma Tubeimoside I(TBMS Ⅰ)on apoptosis of breast cancer cell line MCF-7 by activating Egr1/p21 signaling pathway.Methods:MCF-7 cells were cultured in vitro and divided into blank group,TBMS Ⅰ high-dose group(20 μg/ml),TBMS Ⅰ medium-dose group(15 μg/ml),TBMS Ⅰ low-dose group(10 μg/ml).MCF-7 cells were treated with the corresponding concentration of TBMS Ⅰ.After 24,48 and 72 h of intervention,CCK-8 method was used to detect cell proliferation.After intervention of MCF-7 cells for 48 h,the cell apoptosis and cell cycle distribution in each group were detected by flow cytometry.Western blotting and qRT-PCR were used to detect the protein and mRNA expressions of Egr1 and p21 in each group.Results:After 24,48 and 72 h of intervention,MCF-7 cells in TBMSⅠ high dose group and TBMSⅠ medium dose group were inhibited,and the inhibition rate was time-dependent and dose-dependent.After 48 h of intervention,the apoptosis rate of MCF-7 cells in TBMS Ⅰ high-dose group,TBMS Ⅰ medium-dose group were higher than that in the blank group,difference statistically significant(all P<0.05).The proportion of G0/G1 phase cells in TBMS Ⅰ high-dose group,TBMS Ⅰ medium-dose group and TBMS Ⅰ low-dose group were lower than those in the blank group,and G2/M phase cells higher than that in the blank group,difference statistically significant(all P<0.05).Expression of Egr1 and p21 protein and mRNA in TBMS Ⅰ high-dose group,TBMS Ⅰ medium-dose group and TBMS Ⅰ low-dose group higher than those in the blank group,difference statistically significant(all P<0.05).Conclusion:Tubeimoside I can activate Egr1/p21 signaling pathway in MCF-7 cells,increase the expression of Egr1 and p21,cause G2/M cell cycle arrest,inducing apoptosis of MCF-7 cells.
