摘要:
目的:探讨清热养阴除湿丸对痛风大鼠的治疗机制。方法:将50只SD大鼠随机分为空白组、模型组、秋水仙碱组、清热养阴除湿低剂量组、清热养阴除湿高剂量组。采用改良Coderre 法构建痛风大鼠模型,按组别给予大鼠0.9%氯化钠溶液、秋水仙碱、清热养阴除湿丸灌胃治疗。通过HE染色观察大鼠滑膜组织病理改变; 采用ELISA法检测大鼠外周血IL-1β、IL-18、IL-33、TNF-α水平; 采用RT-qPCR、Western blotting检测大鼠滑膜组织中NLRP3炎症小体相关基因及线粒体自噬相关基因表达。结果:模型组大鼠外周血IL-1β、IL-18、IL-33、TNF-α高于空白组(均P<0.05)。干预后,秋水仙碱组、清热养阴除湿低剂量组、清热养阴除湿高剂量组大鼠外周血IL-1β、IL-18、IL-33、TNF-α下降,且清热养阴除湿高剂量组各项指标最低(P<0.05)。清热养阴除湿低剂量组、清热养阴除湿高剂量组大鼠滑膜组织中NLRP3、Caspase-1、IL-1β、LC3B、p62、PHB2蛋白相对表达低于模型组(均P<0.05)。结论:清热养阴除湿丸通过抑制NLRP3炎症小体形成及线粒体相关自噬的发生减轻痛风大鼠滑膜组织炎症,改善关节功能。
Abstract:
Objective:To explore therapeutic mechanism of Qingre Yangyin Chushi Wan on gout rats.Methods:Fifty SD rats were randomly divided into blank group,model group,colchicine group,Qingre Yangyin Chushi low-dose group and Qingre Yangyin Chushi high-dose group.The gout rat model were constructed using improved coderre method,and given 0.9% sodium chloride solution,colchicine,Qingre Yangyin Chushi Wan by intragastric administration.The pathological changes of synovial tissue were observed by HE staining.The levels of IL-1β,IL-18,IL-33,TNF-α in peripheral blood of rats were detected by ELISA.RT-qPCR and Western blotting were used to detect expression of NLRP3 inflammator-related genes and mitochondrial autophagy related genes in the synovial tissues of rats.Results:IL-1β,IL-18,IL-33 and TNF-α in peripheral blood of rats in the model group were higher than those in the blank group(all P<0.05).After intervention,IL-1β,IL-18,IL-33 and TNF-α in peripheral blood of rats in the colchicine group,Qingre Yangyin Chushi low-dose group and Qingre Yangyin Chushi high-dose group were decreased,and the indexes in Qingre Yangyin Chushi high-dose group were the lowest(all P<0.05).The protein expressions of NLRP3,Caspase-1,IL-1β,LC3B,p62 and PHB2 in synovial tissue of rats in Qingre Yangyin Chushi low-dose group and Qingre Yangyin Chushi high-dose group were lower than those in the model group(all P<0.05).Conclusion:Qingre Yangyin Chushi Wan can alleviate synovial tissue inflammation and improve joint function in gout rats by inhibiting the formation of NLRP3 inflammasomes and occurrence of mitochondrial related autophagy.
