摘要:
目的:探讨益心附葶饮对慢性心力衰竭心肾阳虚证大鼠的影响及其作用机制。方法:结扎左冠状动脉制备大鼠心力衰竭模型,将造模成功的60只大鼠随机分为假手术组、模型组、缬沙坦组(缬沙坦1.14 mg/kg)、中药低剂量组(生药0.25 g/ml)、中药高剂量组(生药1 g/ml)。采用HE染色观察心肌病理组织学变化。各组大鼠干预4周后,ELISA检查血清N端前脑钠肽(NT-proBNP)、超敏C反应蛋白(hs-CRP)、内皮素(ET-1)、白介素6(IL-6),qPCR检测心肌组织HIF-1 mRNA表达,Western blotting检测心肌组织HIF-1α蛋白表达。结果:益心附葶饮可降低心力衰竭大鼠心肌纤维组织表达面积。中药高剂量组大鼠心脏HIF-1α蛋白、mRNA表达高于模型组,差异有统计学意义(均P<0.05)。益心附葶饮还降低心力衰竭大鼠血清NT-proBNP、hs-CRP、ET-1、IL-6水平。 结论:益心附葶饮通过下调HIF-1α蛋白表达,抑制HIF-1/VEGF信号通路介导的抑制炎症反应,降低心力衰竭大鼠血清NT-proBNP、hs-CRP、ET-1、IL-6表达,减缓心力衰竭大鼠心室重构进程。
Abstract:
Objective:To investigate effect and mechanism of Yixin Futing Yin on rats with chronic heart failure with Xinshen Yangxu syndrome.Methods:Sixty rats successfully modeled the left coronary artery were randomly divided into sham surgery group,model group,valsartan group(valsartan 1.14 mg/kg),traditional Chinese medicine low-dose group(crude drug 0.25 g/ml),and traditional Chinese medicine high-dose group(crude drug 1g/ml).Myocardial histopathological changes were observed by HE staining.The rats in each group were treated for 4 weeks,ELISA used to detect serum N-terminal forebrain natriuretic peptide(NT-proBNP),hypersensitivity C-reactive protein(hs-CRP),endothelin(ET-1),interleukin 6(IL-6),qPCR detected expression of HIF-1 mRNA in myocardial tissue,Western blotting detected expression of HIF-1α protein in myocardial tissue.Results:Yixin Futing Yin could reduce the expression area of myocardial fiber tissue in rats with heart failure.The expressions of HIF-1α protein and mRNA in traditional Chinese medicine high-dose group were higher than those in the model group,difference statistically significant(all P<0.05).Yixin Futing Yin also decreased the levels of serum NT-proBNP,hs-CRP,ET-1 and IL-6 in rats with heart failure.Conclusion:Yixin Futing Yin may be mediated by downregulating HIF-1α protein expression and inhibiting HIF-1/VEGF signaling pathway-mediated inflammatory response,and slowing down the ventricular remodeling process of heart failure rats by reducing expression inhibition of serum NT-proBNP,hs-CRP,ET-1 and IL-6 in rats with heart failure.
