摘要:
目的:研究丹参-川芎药对干预血管性痴呆大鼠学习记忆能力的机制,及对海马神经元内的[Ca2+]i、CaM、CaMPKⅡ活性的影响。方法:采用反复夹闭颈动脉、注射硝普钠降压、单侧永久性结扎颈动脉技术建立模型,采用水迷宫定位航行试验和空间探索试验测试大鼠在造模前、造模后、干预后的学习记忆能力,观察VD大鼠海马神经细胞[Ca2+]i变化特征,检测海马组织中CaM、CaMPKⅡ的mRNA水平。结果:丹参-川芎高剂量组与尼莫地平组比较,差异无统计学意义(P>0.05)。丹参-川芎高剂量组穿越原平台区次数与尼莫地平组比较,差异无统计学意义(P>0.05),但优于丹参-川芎低剂量组(P<0.01)。模型组[Ca2+]i荧光强度高于其他各组(均P<0.01),而尼莫地平组、丹参-川芎高剂量组的[Ca2+]i荧光强度比较,差异无统计学意义(均P>0.05)。模型组的CaM mRNA、CaMPKⅡ mRNA含量低于其他各组(均P<0.01),丹参-川芎高剂量组与尼莫地平组比较,差异无统计学意义(P>0.05)。结论:丹参-川芎药对能够阻止VD大鼠海马神经细胞[Ca2+]i过度升高,且减少CaM、CaMPKⅡmRNA表达,能有效改善VD大鼠临床症状。
Abstract:
Objective:To study effects of Danshen Chuanxiong couplet medicines on learning and memory abilities of vascular dementia rats and activities of [Ca2+] i,CaM,and CaMPKⅡ in hippocampal neurons.Methods:Model were established by repeatedly clamping carotid artery,injecting nitroprusside to reduce blood pressure,and unilateral permanent ligation of carotid artery.The learning and memory abilities of rats were tested before,after,and after modeling using a water maze navigation test and spatial exploration test.The changes in [Ca2+] i in hippocampal seminal cells of VD rats were observed,and mRNA levels of CaM and CaMPKⅡ in hippocampal tissue were detected.Results:There was no statistical difference between the high-dose Danshen Chuanxiong group and the Nimodipine group(P>0.05).There was no statistically significant difference in the number of times the high-dose Danshen Chuanxiong group crossed original platform area compared to the Nimodipine group(P>0.05).But it is superior in the low-dose Danshen Chuanxiong group(P<0.01).In terms of [Ca2+] i fluorescence intensity,the model group was significantly higher than other groups(all P<0.01).There were no difference compared to the Nimodipine group,and the high-dose Danshen Chuanxiong group(P>0.05).The levels of CaM mRNA and CaMPKⅡ mRNA in the model group were significantly lower than those in other groups(all P<0.01).There was no difference between the high-dose Danshen Chuanxiong group and the Nimodipine group(P>0.05).Conclusion:Danshen Chuanxiong couplet medicines successfully prevented the excessive elevation of [Ca2+] i in hippocampal neurons of VD rats and reduced expression of CaM and CaMPKⅡ mRNA,while effectively improving clinical symptoms of VD rats.
