摘要:
目的:观察泽兰合剂对肢体缺血再灌注损伤(LIRI)模型大鼠TNF-α、HMGB1表达的影响,探讨泽兰合剂对LIRI炎性损伤的治疗作用。方法:75只雄性SD大鼠,按随机数字表分为五组,对照组、模型组、泽兰合剂低剂量组、泽兰合剂中剂量组、泽兰合剂高剂量组,每组15只,其中按再灌注后不同时间点分为0、2、6 h 3个时间亚组,每组5只。造模后HE染色观察骨骼肌病理组织形态,免疫荧光染色检测骨骼肌肿瘤坏死因子-α(TNF-α)、高迁移率族蛋白B1(HMGB1)表达。ELISA检测血清TNF-α、HMGB1水平变化,RT-qPCR检测骨骼肌中TNF-α、HMGB1的mRNA表达。结果:模型组、泽兰合剂低剂量组、泽兰合剂中剂量组、泽兰合剂高剂量组TNF-α、HMGB1表达较对照组升高,差异有统计学意义(均P<0.05)。泽兰合剂低剂量组、泽兰合剂中剂量组TNF-α、HMGB1含量在相同时间点较模型组降低,其中泽兰合剂中剂量组水平最低,而泽兰合剂高剂量组HMGB1、TNF-α在同时间点较模型组升高,差异有统计学意义(均P<0.05)。结论:中低剂量泽兰合剂均能降低缺血再灌注损伤大鼠TNF-α、HMGB1水平,减轻炎症损伤,且以中剂量泽兰合剂效果最佳,高剂量泽兰合剂则会加剧炎性损伤。
Abstract:
Objective:To observe treatment effects of inflammatory factors of rats with limb ischemia-reperfusion injury(LIRI),and investigate treatment of Zelan mixture for LIRI.Methods:Seventy-five male rats were randomly divided into five groups,control group,model group,low doses of Zelan mixture group,medium doses of Zelan mixture group and high doses of Zelan mixture group,15 rats in each group.Based on the time points of 0 h,2 h and 6 h,subgroups were carried out 5 rats in each group.After establishing limb ischemia reperfusion injury model,the pathological tissue morphology of skeletal muscles were observed by HE staining.The expression of TNF-α,HMGB1 in skeletal muscles were detected by immunofluorescence staining.The expression levels of TNF-α and HMGB1 in serum by ELISA and mRNA levels of TNF-α and HMGB1 in skeletal muscle tissues by RT-qPCR were carried out.Results:The expression of TNF-α and HMGB1 in the model group,the Zelan mixture low dose group,Zelan mixture medium dose group and Zelan mixture high dose group were higher than those in the control group,difference statistically significant(all P<0.05).The contents of TNF-α and HMGB1 in the Zelan mixture low dose group and Zelan mixture medium dose group were lower than those in the model group at the same time point,and levels of HMGB1 and TNF-α in the Zelan mixture high dose group were higher than those in model group at the same time point,with statistical significance(all P<0.05).Conclusion:The levels of TNF-α and HMGB1 in rats with ischemia-reperfusion injury could be reduced by Zelan mixture at medium and low dose,and medium dose Zelan mixture was better.High dose Zelan mixture can aggravate inflammatory injury.
