摘要:
目的:观察益血降浊法方药(YJD)对肾纤维化的保护作用及机制。方法:建立腺嘌呤诱导的小管间质肾病(TIN)大鼠模型和单侧输尿管梗阻(UUO)大鼠模型。TIN大鼠模型组随机分为对照组、TIN模型组、YJD治疗组(低、中、高剂量组)以及依那普利治疗组。UUO大鼠模型组随机分为假手术组、UUO模型组和YJD治疗组,采集各组大鼠血清和肾脏组织,并进行免疫组化分析。结果:YJD改善了TIN大鼠血清肌酐、尿素和尿酸水平,且呈剂量依赖性。UUO大鼠梗阻肾中TGF-β1表达增加,同时p-Smad2、Smad2、p-Smad3、Smad4蛋白表达显著上调,Smad7蛋白表达显著下调。YJD治疗显著抑制了UUO大鼠梗阻肾中TGF-β1的表达,与未治疗的UUO大鼠比较,YJD治疗仅显著抑制UUO大鼠梗阻肾中p-Smad2和p-Smad3蛋白表达,未显示明显逆转UUO大鼠阻塞肾脏中其他蛋白质的表达。与未治疗的UUO大鼠比较,YJD治疗显著抑制了UUO大鼠梗阻肾脏中p-Smad2和p-Smad3核蛋白的表达。组织学上,YJD改善了UUO大鼠的肾纤维化。结论:YJD调节TGF-β1/Smad信号通路,从而改善了肾纤维化。
Abstract:
Objective:To observe the protective effect and mechanism of Yixue Jiangzhuo method (YJD) on renal fibrosis.Methods:Rat models of adenine-induced tubular interstitial nephropathy (TIN) and unilateral ureteral obstruction (UUO) were established.The TIN rat model group was randomly divided into control group,TIN model group,YJD treatment group (low,medium and high-dose groups) and enalapril treatment group.The UUO rat model group was randomly divided into sham operation group,UUO model group and YJD treatment group.Serum and renal tissue of rats in each group were collected and immunohistochemical analysis was performed.Results:YJD improved serum creatinine,urea and uric acid levels in TIN rats in a dose-dependent manner.The expression of TGF-β1 was increased in the obstructive kidney of UUO rats,while the expression of p-Smad2,Smad2,p-Smad3,Smad4 proteins was significantly up-regulated,and the expression of Smad7 protein was significantly down-regulated.YJD treatment significantly inhibited the expression of TGF-β1 in the obstructive kidney of UUO rats.Compared with untreated UUO rats,YJD treatment only significantly inhibited the expression of p-Smad2 and p-Smad3 proteins in the obstructive kidney of UUO rats.Compared with untreated UUO rats,YJD treatment significantly inhibited the expression of p-Smad2 and p-Smad3 nuclear proteins in the obstructive kidney of UUO rats.Histologically,YJD improved renal fibrosis in UUO rats.Conclusion:YJD regulates TGF-β1/Smad signaling pathway,thereby improving renal fibrosis.
