摘要:
目的:探究玉屏风散对变应性鼻炎大鼠肥大细胞表达、免疫应答及NLRP3炎性小体的影响。方法:选取40只大鼠,随机分为正常组、模型组、糠酸莫米松组、玉屏风散组,每组10只,对模型组、糠酸莫米松组、玉屏风散组采用卵清蛋白致敏法建立变应性鼻炎模型,正常组不建立该模型,建模成功后,对糠酸莫米松组给予1喷/侧的糠酸莫米松喷鼻治疗,对玉屏风散组灌胃24 g/kg的玉屏风散,正常组、模型组同期灌胃同体积0.9%氯化钠溶液,对大鼠进行过敏症状进行评分,HE染色法及甲苯胺蓝染色法检测鼻黏膜组织病理形态及肥大细胞表达,ELISA法检测大鼠血清中免疫应答相关因子水平,免疫印迹法检测鼻黏膜组织中NLRP3炎性小体相关蛋白表达。结果:与正常组相比,模型组过敏症状评分、鼻黏膜中肥大细胞数目、OVA-sIgE、IL-4含量、鼻黏膜组织中NLRP3、Caspase-1、ASC蛋白表达升高(P<0.05),血清中INF-γ含量显著降低(P<0.05),与模型组比较,糠酸莫米松组、玉屏风散组过敏症状评分、鼻黏膜中肥大细胞数目、OVA-sIgE、IL-4含量、鼻黏膜组织中NLRP3、Caspase-1、ASC蛋白表达降低(P<0.05),血清中INF-γ含量显著升高(P<0.05),且玉屏风散组比糠酸莫米松组变化显著(P<0.05);与模型组相比,糠酸莫米松组、玉屏风散组病理状态明显改善。结论:玉屏风散对变应性鼻炎大鼠具有显著效果,可有效调控变应性鼻炎大鼠免疫应答、降低肥大细胞表达,并抑制NLRP3炎性小体。
Abstract:
Objective:To investigate the effects of Yupingfeng powder on mast cell expression,immune response and NLRP3 inflammasome in rats with allergic rhinitis.Methods:Forty rats were randomly allocated to four groups (n=10 per group):normal,model,mometasone furoate,and Yupingfeng powder.Allergic rhinitis was induced in the model,mometasone furoate,and Yupingfeng powder groups via ovalbumin sensitization,while the normal group remained untreated.Following successful model establishment,the mometasone furoate group received one spray per nostril of mometasone furoate nasal spray daily.The Yupingfeng powder group was administered 24 g/kg of Yupingfeng powder via oral gavage.Concurrently,the normal and model groups received equivalent volumes of saline.Allergic symptom scores were recorded for all animals.Histopathological evaluation of nasal mucosa and mast cell detection were performed using HE staining and toluidine blue staining,respectively.Serum levels of immune-related factors were measured by ELISA,and Western blot analysis was used to determine the expression of NLRP3 inflammasome-associated proteins in nasal mucosal tissues.Results:Compared to the normal group,the model group exhibited significant increases in allergic symptom scores,mast cell count in nasal mucosa,serum levels of OVA-sIgE and IL-4,as well as protein expression of NLRP3,Caspase-1,and ASC in nasal mucosa tissue (P<0.05),along with a marked reduction in serum INF-γ level (P<0.05).Relative to the model group,both the mometasone furoate and Yupingfeng powder groups showed reductions in allergic symptom scores,nasal mucosal mast cell numbers,OVA-sIgE and IL-4 concentrations,and NLRP3 inflammasome-related protein expressions (P<0.05),while demonstrating a significant elevation in serum INF-γ (P<0.05).Furthermore,the improvements in the Yupingfeng powder group were significantly more pronounced than those in the mometasone furoate group (P<0.05).Histopathological evaluation also indicated substantial amelioration of nasal mucosal lesions in both treatment groups compared to the model group.Conclusion:Yupingfeng powder can effectively regulate the immune response,reduce mast cell expression and inhibit NLRP3 inflammasome in rats with allergic rhinitis.
