摘要:
目的:探究百合多糖对哮喘大鼠气道炎症的改善作用及机制。方法:建立大鼠哮喘模型,随机分为对照组、模型组、百合多糖低、高剂量(150、300 mg/kg)组和地塞米松(1 mg/kg)组,每组12只。灌胃2周后,观察哮喘行为并评分;吉姆萨染色检测支气管肺泡灌洗液(BALF)中的炎症细胞数;ELISA法检测血清免疫球蛋白E(IgE)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平;HE染色和PAS染色观察肺组织病理变化;免疫组化法检测肺组织Toll样受体4(TLR4)蛋白表达;Western blot检测肺组织磷脂酰肌醇3-激酶/蛋白激酶B通路(PI3K/Akt)通路和核因子κB p65(NF-κB p65)活化情况。结果:与对照组比较,模型组大鼠哮喘症状评分,BALF中炎症细胞数量,血清IgE、IL-6、TNF-α水平升高(均P<0.05);肺组织呈现典型哮喘病理改变,且炎症及PAS评分、TLR4、p-NF-κB p65/NF-κB p65、p-PI3K/PI3K、p-Akt/Akt蛋白水平升高(均P<0.05)。与模型组比较,百合多糖低、高剂量组和地塞米松组大鼠哮喘症状评分,BALF中炎症细胞数量,血清IgE、IL-6、TNF-α水平降低(均P<0.05);肺组织哮喘病理改变程度均减轻,且炎症及PAS评分、TLR4、p-NF-κB p65/NF-κB p65、p-PI3K/PI3K、p-Akt/Akt蛋白水平降低(均P<0.05);且百合多糖的作用呈剂量依赖性(均P<0.05)。结论:百合多糖可减轻哮喘大鼠气道炎症及黏液高分泌,改善哮喘症状,其机制可能与抑制PI3K/Akt/NF-κB信号通路有关。
Abstract:
Objective:To investigate the ameliorative effect and underlying mechanism of Lilium polysaccharide on airway inflammation in asthmatic rats.Methods:A rat asthma model was established and randomly divided into a control group,a model group,low- and high-dose Lilium polysaccharide groups (150,300 mg/kg),and a dexamethasone group (1 mg/kg),with 12 rats in each group.Drugs were administered by gavage for 2 weeks.Asthma-like behaviors were observed and scored.Inflammatory cells in BALF were counted with Giemsa staining.Serum IgE,IL-6 and TNF-α were measured by ELISA.Lung histopathology was evaluated with HE and PAS staining.TLR4 protein expression was detected by immunohistochemistry.And activation of PI3K/Akt pathway and NF-κB p65 was determined by Western blot.Results:Compared with the control group,in the model group,the asthma symptom score of rats,the number of inflammatory cells in bronchoalveolar lavage fluid (BALF),and the levels of serum IgE,IL-6,and TNF-α were increased (all P<0.05).The lung tissue showed typical pathological changes of asthma,and the inflammation and PAS scores,as well as the protein levels of TLR4,p-NF-κB p65/NF-κB p65,p-PI3K/PI3K,and p-Akt/Akt were increased (all P<0.05).Compared with the model group,the asthma symptom scores of rats in the low-and high-dose groups of Lilium polysaccharide and the dexamethasone group,the number of inflammatory cells in BALF,and the levels of serum IgE,IL-6,and TNF-α were decreased (all P<0.05).The degree of pathological changes of asthma in lung tissue was alleviated,and the inflammation and PAS scores,and the protein levels of TLR4,p-NF-κB p65/NF-κB p65,p-PI3K/PI3K and p-Akt/Akt were decreased (all P<0.05).Moreover,the effects of Lilium polysaccharide were dose-dependent (all P<0.05).Conclusion:Lilium polysaccharide can attenuate airway inflammation and mucus hyper-secretion,improve asthmatic symptoms and relieve lung pathological injury in asthmatic rats,possibly by inhibiting the PI3K/Akt/NF-κB signaling pathway.
