摘要:
溃疡性结肠炎(UC)是一种由黏膜屏障受损、免疫炎症失衡、肠道菌群紊乱及胆汁酸代谢异常等多因素驱动的慢性炎症性肠病。法尼醇X受体(FXR)是维持“肠肝轴”稳态的胆汁酸核受体,通过调控胆汁酸合成与转运、增强肠黏膜屏障功能及抑制炎症反应,在UC发生发展中发挥重要调节作用。中医药在整体调控“胆汁酸-肠黏膜-肠道菌群”方面具有多成分、多靶点优势。笔者在阐述FXR信号通路生物学功能的基础上,系统综述近年中药有效成分、单味中药及复方以FXR为关键靶点干预UC的实验研究进展,归纳其在重建胆汁酸稳态、修复黏膜屏障和重塑肠道微生态方面的作用特点,为中医药基于FXR信号通路防治UC提供理论依据。
Abstract:
Ulcerative colitis (UC) is a chronic inflammatory bowel disease driven by multiple factors,including disruption of the mucosal barrier,immune dysregulation,gut microbiota dysbiosis,and disordered bile acid metabolism.Farnesoid X receptor (FXR),a bile acid-activated nuclear receptor,is pivotal for maintaining gut-liver axis homeostasis.By coordinating bile acid synthesis and transport,reinforcing intestinal barrier integrity,and suppressing inflammatory signaling,FXR critically modulates the pathogenesis and progression of UC.Traditional Chinese medicine (TCM) offers multicomponent and multitarget advantages and can holistically regulate the bile acid-mucosa-microbiota network.This review summarizes the biological functions of FXR signaling and synthesizes recent experimental evidence on FXR-targeted TCM interventions for UC,including purified active constituents,single herbs,and compound formulas.We highlight their shared actions in restoring bile acid homeostasis,repairing the mucosal barrier,and reshaping the gut microbial ecosystem,thereby providing a theoretical basis for FXR-centered TCM strategies for UC prevention and treatment.
