摘要:
目的:观察二陈汤对Lewis肺癌小鼠的抑瘤率、血常规,以及肿瘤组织中Gli-1、SHH、SMO、PTCH-1等蛋白表达的影响。方法:用40只C57BL/6小鼠建立Lewis肺癌小鼠模型,造模成功后随机分为模型组、中药组、联合组以及顺铂组,每组各10只。7 d后中药组、联合组给予二陈汤灌胃,模型组给予同等量0.9%氯化钠溶液。顺铂组给予注射用顺铂溶液腹腔内注射,同时予以0.9%氯化钠溶液灌胃,连续14 d。观察小鼠的一般情况,肿瘤生长情况。21 d后处死,摘眼取血并剥离肿瘤组织。常规生化法检测血常规水平; 免疫组化法、Western blot法检测肿瘤组织中SMO、Gli-1、PTCH-1、SHH蛋白表达水平。结果:中药组与联合组均能改善小鼠的肿瘤生长、提高抑瘤率,联合组相比其他各组抑瘤率更高,但与顺铂组比较差异无统计学意义(P>0.05)。各用药干预后顺铂组、联合组小鼠WBC、RBC计数较模型组和中药组均降低(P<0.05),联合组血红蛋白含量较顺铂组增加(P<0.05)。中药组和联合组可不同程度抑制小鼠肿瘤组织中SMO、Gli-1、PTCH-1、SHH的蛋白表达(均P<0.05)。结论:二陈汤联合顺铂可以显著抑制Lewis小鼠肿瘤增殖和克隆形成能力,降低小鼠瘤组织中SHH、SMO、PTCH-1和Gli-1蛋白表达水平。
Abstract:
Objective:To observe the effect of Erchen decoction on the expression of Hedgehog pathway associated protein in Lewis lung cancer mice.Methods:40 C57BL/6 mice were used to establish the lung cancer model.When the model was finished,these mice were randomly divided into four groups,with ten mice in each group.7 days later,the traditional Chinese medicine(TCM)group and the combined group were given the Erchen decoction by stomach irrigation,as the model group was assigned the same amount of normal saline.Cisplatin group was injected intraperitoneally with cisplatin solution and irrigated stomach with normal saline for 14 days.Mice's general condition and tumor growth of mice were observed.After 21 days,collect mice blood from retro-orbital plexus for the blood test and the axillary tumor tissue; the expression levels of SMO,Gli-1,PTCH-1,and SHH were detected by IHC and Western blot.Results:Both combined and cisplatin groups can suppress tumor growth and inhibition rates.The combined group tumor inhibition rate is higher than other groups,but there is no significance when compared with the cisplatin group(P<0.05).The WBC and RBC figures of the cisplatin group,the combined group were decreased compared with model and TCM group(P<0.05),and hemoglobin count in the combined group increased compared with cisplatin group(P<0.05).The TCM group,the combined group could inhibit the expression of Hedgehog pathway protein in mouse tumor tissues to varying degrees(all P<0.05).Conclusion:Erchen decoction combined with cisplatin can inhibit the tumor proliferation and clonogenic ability of Lewis mice and reduce SHH,SMO,PTCH-1,and Gli-1 protein expression levels in mouse tumor tissues.
