摘要:
目的:基于Th17/Treg细胞平衡,探究整体调节针法对骨质疏松症(OP)大鼠的调控作用。方法:30只大鼠随机分为假手术组、模型组、整体调节针法组。行双侧卵巢切除术构建OP模型,整体调节针法组大鼠针刺足三里、肾俞、大杼,并在双侧肾俞进行温和灸治疗3 个月。干预结束后,Micro-CT评估大鼠股骨形态学参数:骨密度(BMD)、骨体积分数(BV/TV)、骨小梁数量(Tb.N)、骨小梁分离度(Tb.Sp)和骨小梁厚度(Tb.Th)的变化;流式细胞术定量脾脏 Th17和Treg细胞比例;ELISA检测血清中碱性磷酸酶(ALP)、Ⅰ型前胶原N-端前肽(PⅠNP)、抗酒石酸酸性磷酸酶 5b(TRACP-5b)、Ⅰ型胶原 C-端肽(CTX-Ⅰ)的水平。结果:与假手术组比较,模型组BMD、BV/TV、Tb.N、Tb.Th降低,Tb.Sp升高(均P<0.05)。骨代谢ALP、PⅠNP降低,TRACP-5b、CTX-Ⅰ升高(均P<0.05)。Th17比例、Th17/Treg比值升高,Treg比例降低(P<0.05)。整体调节针法组较模型组显著逆转了上述异常指标(P<0.05)。结论:整体调节针法通过改善骨微结构、调节骨代谢标志物、纠正Th17/Treg失衡,发挥抗OP作用。
Abstract:
Objective:In the context of Th17/Treg cell balance,this study aimed to explore impact of the holistic regulatory acupuncture therapy on rats with osteoporosis (OP).Methods:30 rats were randomly and equally divided into three groups:the sham-operated group,the model group,and the holistic regulatory acupuncture group.The OP model was established through bilateral ovariectomy.In the holistic regulatory acupuncture group,rats received acupuncture at Zusanli,Shenshu,and Dazhu.Additionally,mild moxibustion treatment was administered at bilateral Shenshu for a duration of three months.Upon completion of the intervention,Micro-CT was employed to assess the morphological parameters of the rat femur,including bone mineral density (BMD),bone volume fraction (BV/TV),trabecular number (Tb.N),trabecular separation (Tb.Sp),and trabecular thickness (Tb.Th).Flow cytometry was utilized to quantitatively analyze the proportions of Th17 and Treg cells in the spleen.Enzyme-linked immunosorbent assay (ELISA) was used to measure the levels of alkaline phosphatase (ALP),type Ⅰ procollagen N-terminal propeptide (PⅠNP),tartrate-resistant acid phosphatase 5b (TRACP-5b),and type Ⅰ collagen C-terminal telopeptide (CTX-Ⅰ) in the serum.Results:Compared with the sham-operated group,the model group exhibited a decrease in BMD,BV/TV,Tb.N,and Tb.Th,along with an increase in Tb.Sp (all P<0.05).Regarding bone metabolism,the levels of ALP and PⅠNP decreased,while those of TRACP-5b and CTX-Ⅰincreased (all P<0.05).Moreover,in the model group,the proportion of Th17 cells and the Th17/Treg ratio increased,while the proportion of Treg cells decreased (P<0.05).In contrast,the holistic regulatory acupuncture group significantly reversed these abnormal indicators compared to the model group (P<0.05).Conclusion:The holistic regulatory acupuncture therapy exerts an anti-OP effect by improving bone microstructure,regulating bone metabolism markers,and rectifying the Th17/Treg imbalance.
