摘要:
目的:探讨健脾养阴祛瘀方对胃癌前病变(PLGC)大鼠IL-6/JAK2/STAT3通路调控胃黏膜上皮-间质转化(EMT)的作用机制。方法:58只SD大鼠随机分为空白对照组和模型建立组,采用多因素复合方法建立PLGC大鼠模型。造模成功后将模型建立组分为模型组、叶酸组及健脾养阴祛瘀方高、中、低剂量组,每组8只。治疗8周后,采用HE染色观察胃组织病理变化,ELISA检测TNF-α、IL-6水平,qPCR检测JAK2、STAT3的mRNA表达;Western Blot检测JAK2/p-JAK2和STAT3/p-STAT3蛋白表达;免疫组化检测C-myc蛋白表达。结果:健脾养阴祛瘀方能改善胃黏膜萎缩、肠化生、异型增生等病理表现,下调IL-6、TNF-α、JAK2、p-JAK2、STAT3、p-STAT3和C-myc表达(P<0.05),其中健脾养阴祛瘀方高剂量组效果较好。结论:健脾养阴祛瘀方通过改善炎症微环境,抑制JAK2/STAT3通路活化和C-myc表达,抑制EMT进程,缓解PLGC。
Abstract:
Objective:To investigate mechanism by Jianpi Yangyin Quyu formula regulates gastric mucosal epithelial-mesenchymal transition (EMT) via the IL-6/JAK2/STAT3 pathway in rats with precancerous lesions of gastric cancer (PLGC).Methods:A total of 58 SD rats were randomly divided into blank control group and model establishment group.The PLGC rat model was induced using a multi-factor composite method.After successful modeling,the model establishment group were further divided into model group,folic acid group,and high-dose,medium-dose and low-dose Jianpi Yangyin Quyu formula formula groups (n=8 per group).After 8 weeks of treatment,HE staining was used to observe pathological changes in gastric tissue.ELISA was performed to detect TNF-α and IL-6 levels.qPCR was used to measure mRNA expression of JAK2 and STAT3.Western blotting was employed to assess the protein expression of JAK2/p-JAK2 and STAT3/p-STAT3.Immunohistochemistry was applied to evaluate C-myc protein expression.Results:Jianpi Yangyin Quyu formula formula significantly ameliorated pathological manifestations,including gastric mucosal atrophy,intestinal metaplasia,and dysplasia.It down regulated the expression of IL-6,TNF-α,JAK2,p-JAK2,STAT3,p-STAT3,and C-myc (P<0.05),with the high-dose Jianpi Yangyin Quyu formula group exhibiting the most pronounced effects.Conclusion:Jianpi Yangyin Quyu formula formula alleviates PLGC by improving inflammatory microenvironment,inhibiting the activation of the JAK2/STAT3 pathway and C-myc expression,and suppressing the EMT process.
